Diagnostisessa patologian laboratoriossa on k\u00e4yt\u00f6ss\u00e4 satoja vasta-aineita. Immunohistokemiallisen vasta-aineen toiminta diagnostiikassa on ennen\nk\u00e4ytt\u00f6\u00f6n ottamista todistettava ja dokumentoitava, eli verifioitava. T\u00e4ss\u00e4 tutkielmassa luotiin palvelimelle p\u00e4ivitt\u00e4iseksi ty\u00f6kaluksi patologeille ja sairaalasolubiologeille digitaalinen verifiointiarkisto, josta voi tarkistaa kunkin vasta-aineen k\u00e4ytt\u00f6aiheen ja v\u00e4rj\u00e4yspatterin. Digiarkistoon p\u00e4ivitettiin keskeisimm\u00e4t diagnostisten immunohistokemiallisten vasta-aineiden\nverifointiraportit, ja digitoitiin raportteihin kuuluvat normaali- ja tuumorikudosverifiointilasit mikroskooppiskannerin avulla. Verifiointiarkiston rakentumista demonstroitiin t\u00e4ss\u00e4 tutkielmassa verifioimalla aivolis\u00e4kkeen\netulohkon soluista per\u00e4isin olevien rauhaskasvaimien, adenoomien, diagnostiikkaan tarkoitetut uudet transkriptiofaktorivasta-aineet Pit-1, SF-1 ja TPit, joita WHO:n aivolis\u00e4kkeen neuroendokriinisten kasvainten nykyluokitus vaatii t\u00e4sm\u00e4llisen diagnoosin asettamiseksi, ja aivolis\u00e4kkeen hormoneja tuottavien solutyyppien erottamiseksi. SF-1 -vasta-aineen todettiin t\u00e4ss\u00e4\ntutkimuksessa v\u00e4rj\u00e4\u00e4v\u00e4n adenoomia, joiden odotettiin v\u00e4rj\u00e4ytyv\u00e4n vain Pit-1 - vasta-aineella. Hundreds of antibodies are used in the diagnostic pathology laboratory. The function of an immunohistochemical antibody must be proven and documented, in other words verified, before using in the diagnostics. In this thesis, a digital verification archive was created on the server as a daily tool for pathologists and medical cell biologists. The indication and the staining pattern for each antibody can be\nchecked from the archive. The most important diagnostic immunohistochemical antibody verification reports were updated in the digital archive, and the normal and tumor tissue verification glasses included in the reports were digitized using\na microscope scanner. The verification archive was demonstrated in this thesis by verifying the new transcription factor antibodies Pit-1, SF-1 and T-Pit, intended for the diagnosis of glandular tumors (adenomas) originating from the cells of the anterior pituitary lobe. The antibodies are required by the current WHO classification of pituitary neuroendocrine tumors to establish an accurate\ndiagnosis, and to distinguish cell types producing pituitary hormones. In this study, the SF-1 antibody was found to stain adenomas, that were expected to be stained only with the Pit-1 antibody. There are double-positive Pit-1 and SF-1\nadenomas, but they are uncommon.
