Fytokromiin sitoutunut molekyyli, kromofori, absorboi fotonin ja saa aikaan muutoksia fytokromin kolmiulotteisessa rakenteessa: punaista valoa absorboiva Pr-tila virittyy Pfr-tilaksi, joka palaa takaisin Pr-tilaan absorboimalla kaukopunaista valoa tai ilman valotusta pime\u00e4palautumalla. Fytokromin valovaihtuvuudessa t\u00e4rke\u00e4n rakenteen on havaittu olevan fytokromi-spesifisess\u00e4 domeenissa (PHY-domeenissa) sijaitseva PHY-lis\u00e4ke. T\u00e4ss\u00e4 tutkielmassa pyrkimyksen\u00e4 oli tuottaa ja puhdistaa Deinococcus radiodurans -bakteerifytokromia ilman PHY-lis\u00e4kett\u00e4 ja verrata mutantin UV-Vis-spektri\u00e4 villityypin spektriin. Lis\u00e4ksi selvitettiin, saadaanko mutantti valotettua Pfr-tilaan. Mutanttia saatiin tuotettua ja puhdistettua pieni m\u00e4\u00e4r\u00e4, 0,51 mg. Mutantin valotus Pfr-tilaan onnistui. Mutantin ja villityypin UV-Vis-spektrit erosivat toisistaan, sill\u00e4 mutantti absorboi villityyppi\u00e4 v\u00e4hemm\u00e4n punaista ja kaukopunaista valoa. Chromophore, a molecule bound to phytochrome, absorbs a photon and causes changes in the three-dimensional structure of the phytochrome: the red light absorbing Pr state can be excited to the far-red light absorbing Pfr state. The phytochrome returns to the Pr state by the absorption of far-red light or thermally in dark. An important structure in photoactivation has been located to the phytochrome specific domain (PHY-domain) and the hairpin structure in the phytochrome specific domain. The aim of this thesis was to express and purify bacteriophytochrome of Deinococcus radiodurans without the hairpin structure and to compare the UV-Vis spectra of the mutant with the wild type. In addition, the aim was to find out if the mutant can be excited to the Pfr state. The expression of the mutant was successful and a small amount of phytochrome was purified, 0.51 mg. The mutant could be excited to the Pfr state. The UV-Vis spectra of the mutant and wild type phytochrome were different from one another: the mutant absorbed less red and far-red light than the wild type. In dark reversion experiment, where the kinetics of returning to Pr state from Pfr state was studied, it was observed that the mutant reversed faster than the wild type. The main conclusion is that the hairpin structure in the phytochrome specific domain can be removed while maintaining the ability of the phytochrome to be activated by light.
