Tämän tutkimuksen tavoitteena oli selvittää (1) miten faagit vaikuttavat bakteerin taudinaiheuttamiskykyyn liittyvään pesäkemorfologiaan ja faagiresistenssin kehittymiseen, (2) voidaanko bakteeria infektoida yhtä faagia tehokkaammin faagikoktaililla, joka sisältää yhtä tai kahta faagityyppiä ja (3) voidaanko faageja evolvoida infektoimaan faagiresistenttejä bakteeri-isolaatteja. Tässä työssä eri puolilta Suomea sijaitsevilta kalanviljelylaitoksilta eristettyjä F. columnare -bakteerikantoja altistettiin joko yhdelle tai kahdelle niitä infektoivalle faagille. Käsittelyistä määritettiin bakteeritiheys, pesäkkeiden morfologiajakauma ja faagitiitteri. Kokeen lopussa seulottiin evolvoituneita faageja. Tuloksiksi saatiin, että lähes kaikissa käsittelyissä pesäkkeiden morfologia muuttui, mikä saattaa viitata taudinaiheuttamiskykyisempien bakteerien määrän vähenemiseen. Faagiresistenttiys saattoi usein vaikuttaa bakteerin pintareseptoriin. Kahden faagin käyttäminen yhden sijasta ei voimistanut bakteerin infektointia. Kahta faagityyppiä sisältävässä käsittelyssä havaittiin evolvoituneita faageja. Tulokset tukevat faagien mahdollista hyödyntämistä columnaris-tautia vastaan. Flavobacterium columnare is a Gram-negative bacterium causing columnaris disease. This disease causes economical losses at fish farms and during the last decades the number of outbreaks has increased. The main cure has been antibiotics, but also bacteriophages could be used. The research questions of the study were (1) how presence of phages change bacteria’s colony morphology related to virulence, and development of phage resistance, (2) does phage cocktail of one or two phage types infect bacterium more effectively than one phage, and (3) is it possible to evolve phages to infect phage-resistant bacterial isolates. Bacteria and their phages used in this study were previously isolated from Finnish fish farms. F. columnare bacterium strain was exposed to either one or two phages. From the treatments, bacterium concentration, distribution of bacterium colony morphotype and phage titre were determined. At the end of the experiment, phages which could infect phage resistant bacterium were screened. As a result, almost in all treatments bacterium morphology changed. This might suggest that the number of more virulent bacterium decreased. Phage resistance might often affect the surface receptor of bacterium. Use of two phages was not more effective than one for infecting bacterium. Evolved phages from two phage type treatment were isolated. Results support the scenario of utilizing phages against columnaris disease.
