Viimeisimm\u00e4t tutkimukset osoittavat, ett\u00e4 HDL\ntehostaa glukoosin hapetusta ja vaikuttaa my\u00f6s rasvahappojen hapetukseen\nmitokondrioissa. Ideana oli tutkia HDL:n eri alaluokkien, HDL2:n ja HDL3:n,\nvaikutusta hiilihydraattien ja rasvahappojen oksidaatioon hiiren C2C12-\nluustolihassoluissa. Kokonaisten myoblastien ja permeabilisoitujen myotuubien\nhapenkulutusta mitattiin korkean resoluution respirometri -menetelm\u00e4ll\u00e4 (engl.\nhigh-resolution respirometry). Solut k\u00e4siteltiin ihmisen HDL:ll\u00e4 ja niiden\nhapenkulutusta mitattiin eri energiasubstraattien ja inhibiittorien l\u00e4sn\u00e4ollessa, jotta\nniiden metaboliaa voitiin analysoida. Hiilihydraattimittauksissa myoblasteille\ntarjottiin glukoosia ja myotuubeille mitokondrion kompleksi I:n ja kompleksi II:n\nsubstraatteja. Rasvahappomittauksissa molemmille tarjottiin tietty\u00e4 rasvahappoa.\nHDL2 tehosti rasvahappojen hapetusta myotuubeissa kontrolliin n\u00e4hden. Lis\u00e4ksi\nHDL3 tehosti mitokondrion kompleksin I:n substraattien hapetusta myotuubeissa\nkontrolliin n\u00e4hden. HDL ei kuitenkaan vaikuttanut myoblastien\nhapenkulutukseen. Voidaan todeta, ett\u00e4 HDL n\u00e4ytt\u00e4\u00e4 vaikuttavan rasvahappojen\nhapetukseen sek\u00e4 mitokondrion kompleksin I kautta tapahtuviin\nhapetusprosesseihin myotuubeissa. Fascinatingly, recent studies have\nrevealed that HDL affects beneficially mitochondrial function and subsequently\ncellular energy metabolism. HDL is proven to enhance mitochondrial glucose\noxidation and affect fatty acid oxidation. The aim was to study the effect of human\nHDL subfractions, HDL2 and HDL3, on mitochondrial carbohydrate and fatty acid\noxidation in C2C12 mouse skeletal muscle cells. Intact myoblasts and permeabilized\nmyotubes were measured using a high-resolution respirometry. The cells were\ntreated with HDL and oxygen consumption of the cells was measured in the\npresence of different energy substrates and inhibitors. This allowed evaluation of\nmetabolism of the cells. Mitochondrial complex I and complex II substrates were\noffered to myotubes and glucose to myoblasts in order to analyze carbohydrate\noxidation. Fatty acid oxidation was also evaluated by offering fatty acid substrates\nto the cells. HDL2 increased fatty acid oxidation and HDL3 increased complex I\nsubstrate oxidation in myotubes. Though, no effect in myoblasts was noticed. In\nconclusion, HDL seems to affect fatty acid oxidation and oxidation processes\noccurring via the complex I in myotubes.
