Tämän tutkimuksen tavoitteena oli analysoida FLiP:n ja sen isäntäbakteerien välisiä vuorovaikutuksia. FLiP:n elinkiertoa tarkasteltiin adsorptiotestien ja kasvukäyräkokeiden avulla ja sen proteiineja analysoitiin kemikaalikäsittelyjen, elektroforeesin ja zymografian avulla. Isäntäkirjon evoluutiokokeissa FLiP-faagia lisättiin eristysisännästä ja mahdollisista uusista isännistä koostuviin sekaviljelmiin. Adsorptiotestien nesteviljelmissä FLiP kiinnittyi ainoastaan eristysisäntäänsä, vaikka puolikiinteillä kasvatusalustoilla FLiP kykenee infektoimaan lisäksi myös kahta muuta bakteerikantaa. FLiP:n proteiineista saatiin uutta tietoa. FLiP:llä esimerkiksi havaittiin useita lyyttisiä entsyymejä. Tämä saattaisi viitata endolyysiinien esiintymiseen virionissa sekä mahdollisesti myös faagin laajaan isäntäkirjoon. Mikäli FLiP:n isäntäkirjo laajenisi kaloille patogeenisiin flavobakteerikantoihin, FLiP voisi toimia hyvänä faagiterapian välineenä tappaessaan kohdennetusti ja tehokkaasti kalojen kudoksiin kiinnittyneitä bakteereja. Single-stranded DNA (ssDNA) phages, lipid-containing phages and non-tailed phages are more common and ecologically more significant than previously considered, though still relatively scarcely isolated and researched. FLiP is a Flavobacterium-infecting virus, which has been isolated from the lake Jyväsjärvi in 2010. FLiP has an internal lipid membrane, ssDNA genome and icosahedral, nontailed capsid. The aim of this study was to analyze interactions between FLiP and its host bacteria. Adsorption tests and one-step cultivations were performed to analyse adsorption and infective cycle of FLiP. Chemical treatments, electrophoresis and zymography were used to analyse phage proteins. Mixed cultures of original and potential new hosts were applied to test host-range evolution of FLiP. FLiP can infect three bacterial strains on semi-solid agar, but it could only adsorb onto its original host in liquid cultivations of adsorption tests. New information on FLiP proteins was acquired; for example, several lytic enzymes were observed. That might suggest that FLiP contains endolysins. Several lytic enzymes might also implicate that the phage has broad host range. If the host range of FLiP was broadened to fish-pathogenic Flavobacterium-strains, FLiP could be used as a good agent for phage therapy targeting specifically and effectively the bacteria, which are attached to fish tissues.
