Taudin globaali vallitsevuus on 30 prosenttia. Suolistomikrobisto koostuu ihmiselle elint\u00e4rkeist\u00e4 mikrobeista, jotka vaikuttavat ihmisen terveyteen. Suolisto ja maksa ovat tiiviiss\u00e4 yhteydess\u00e4 porttilaskimon v\u00e4lityksell\u00e4. Suolistomikrobiston koostumus on yhteydess\u00e4 ei-alkoholiper\u00e4iseen rasvamaksaan. T\u00e4m\u00e4n ty\u00f6n tavoitteena oli selvitt\u00e4\u00e4 eroja suolistomikrobiston koostumuksessa ja ravitsemuksessa terveiden ja ei-alkoholiper\u00e4ist\u00e4 rasvamaksaa sairastavien v\u00e4lill\u00e4. Tavoitteena oli my\u00f6s selvitt\u00e4\u00e4, vaikuttaako ei-alkoholiper\u00e4inen rasvamaksa ravitsemuksen ja suolistomikrobiston v\u00e4liseen yhteyteen. Tarkoituksena oli tuottaa lis\u00e4\u00e4 tutkimusn\u00e4ytt\u00f6\u00e4 siit\u00e4, vaikuttaako ravitsemus suolistomikrobiston koostumukseen terveill\u00e4 ja ei-alkoholiper\u00e4ist\u00e4 rasvamaksaa sairastavilla samalla tavalla.\n\nTutkimusaineistona k\u00e4ytettiin Jyv\u00e4skyl\u00e4n yliopiston MAKSA-aineistoa, joka on ker\u00e4tty vuosina 2020\u20132022. Tutkittavat olivat ylipainoisia aikuisia, joista osalla oli ei-alkoholiper\u00e4inen rasvamaksa (n=28) ja osalla ei (n=13). Ei-alkoholiper\u00e4inen rasvamaksa todettiin keskussairaalassa magneettiresonanssikuvauksella. Tutkittavilta otettiin ulosten\u00e4ytteet. Ulosten\u00e4ytteist\u00e4 eristettiin suolistobakteerien DNA, jonka avulla m\u00e4\u00e4ritettiin suolistomikrobiston koostumus. Tutkittavat t\u00e4yttiv\u00e4t ruokap\u00e4iv\u00e4kirjaa kolmen p\u00e4iv\u00e4n ajan. Ravinnonsaanti analysoitiin Aivodiet-ohjelman avulla. Suolistomikrobiston erot analysoitiin CLC Microbial Genomics Package -ohjelmalla. Erot ravitsemuksessa analysoitiin T-testill\u00e4 ja Mann-Whitneyn U-testill\u00e4. Ei-alkoholiper\u00e4isen rasvamaksan vaikutusta ravitsemuksen ja suolistomikrobiston v\u00e4liseen yhteyteen analysoitiin korrelaatioanalyysill\u00e4. \n\nSuolistomikrobistossa eroja l\u00f6ytyi kahdeksassa bakteerien suvussa ja kahdessa bakteerien heimossa terveiden ja ei-alkoholiper\u00e4ist\u00e4 rasvamaksaa sairastavien v\u00e4lill\u00e4. Tilastollisesti merkitsevi\u00e4 eroja l\u00f6ytyi esimerkiksi Herbinix-suvun bakteerien (p=0.04) m\u00e4\u00e4r\u00e4ss\u00e4. Ravitsemuksessa ei havaittu eroja terveiden ja ei-alkoholiper\u00e4ist\u00e4 rasvamaksaa sairastavien v\u00e4lill\u00e4. Terveill\u00e4 ja ei-alkoholiper\u00e4ist\u00e4 rasvamaksaa sairastavilla havaittiin erilaisia korrelaatioita ravitsemusmuuttujien ja suolistobakteerien v\u00e4lill\u00e4. Ei-alkoholiper\u00e4ist\u00e4 rasvamaksaa sairastavilla ei l\u00f6ytynyt yht\u00e4k\u00e4\u00e4n tilastollisesti merkitsev\u00e4\u00e4 korrelaatiota ravitsemusmuuttujien ja suolistobakteerien v\u00e4lill\u00e4. Terveill\u00e4 l\u00f6ytyi tilastollisesti merkitsev\u00e4 korrelaatio kolesterolin saannin ja Herbinix-suvun bakteerien (r=0.61, p=0.03), kuidun saannin ja Lactobacillus-suvun bakteerien (r=0.61, p=0.02) sek\u00e4 kuidun saannin ja Lactobacillaceae-heimon bakteerien (r=0.62, p=0.02) v\u00e4lill\u00e4. \n\nTulokset osoittavat, ett\u00e4 ei-alkoholiper\u00e4ist\u00e4 rasvamaksaa sairastavilla on erilainen suolistomikrobiston koostumus kuin terveill\u00e4. The global prevalence of NAFLD is 26 percent. The gut microbiota consists of essential microbes that affect human health. The gut and liver are closely connected through the portal vein. The aim of this study was to investigate differences in the composition of the gut microbiota and nutrition between healthy individuals and those with NAFLD. Additionally, the aim was to determine whether NAFLD influences the relationship between nutrition and the gut microbiota. The purpose was to provide further research evidence on whether nutrition influences the composition of the gut microbiota in the same way in healthy individuals and those with non-alcoholic fatty liver disease.\nThe study used the MAKSA dataset from the University of Jyv\u00e4skyl\u00e4, collected between 2020 and 2022. The participants were overweight adults, some of whom had NAFLD (n=28), and others did not (n=13). NAFLD was diagnosed at the central hospital using magnetic resonance imaging (MRI). Stool samples were collected from the participants, and the DNA of the gut bacteria was extracted to determine the composition of the gut microbiota. The participants kept a food diary for three days, and their nutrient intake was analyzed using the Aivodiet program. Differences in the gut microbiota were analyzed using the CLC Microbial Genomics Package. Nutritional differences were analyzed using the T-test and Mann-Whitney U-test. The influence of NAFLD on the relationship between nutrition and the gut microbiota was analyzed using correlation analysis.\nDifferences in the gut microbiota were found in eight bacterial genera and two bacterial families between healthy individuals and those with NAFLD. Statistically significant differences were found, for example, in the abundance of bacteria from the genus Herbinix (p=0.04). No differences were observed in nutrition, but the relationships between nutritional variables and gut bacteria were different in healthy individuals and those with NAFLD. In those with NAFLD, no statistically significant correlations were found between nutritional variables and gut bacteria. In healthy individuals, a statistically significant correlation was found between cholesterol intake and Herbinix genus bacteria (r=0.61, p=0.03), fiber intake and Lactobacillus genus bacteria (r=0.61, p=0.02), and fiber intake and Lactobacillaceae family bacteria (r=0.62, p=0.02).\nThe results indicate that individuals with NAFLD have a different gut microbiota composition than healthy individuals. The lack of nutritional differences may be due to the fact that all participants were overweight.
