T\u00e4ss\u00e4 Pro gradu -tutkielmassa tutkittiin eri tapoja lis\u00e4t\u00e4 viljeltyjen ihmislihassolujen proteiinisynteesi\u00e4 sek\u00e4 n\u00e4iden lihassolujen glukoosin k\u00e4ytt\u00f6\u00e4 biomolekyylien synteesiss\u00e4. Viljellyille ihmislihassoluille tehtiin solukokeita tarkoituksena lis\u00e4t\u00e4 lihassolujen proteiinisynteesi\u00e4. Lihassoluista eristettiin proteiinit ja niiden proteiinikonsentraatiot mitattiin. Nestetuikelaskennan avulla tutkittiin lihassolujen sis\u00e4lt\u00e4mien proteiinien m\u00e4\u00e4ri\u00e4 sek\u00e4 glukoosin k\u00e4ytt\u00f6\u00e4 proteiinien, RNA:n ja lipidien synteesiss\u00e4. Seriinisynteesireitill\u00e4 toimivan entsyymin, 3-fosfoglyseraattidehydrogenaasin (eng. Phosphoglycerate dehydrogenase, PHGDH), sek\u00e4 Rapamysiinin mekanistisen kohteen 1 (eng. The mechanistic target of rapamycin complex 1, mTORC1) aktiivisuudesta kertovien fosforyloituneiden S6K1- ja S6-proteiinien m\u00e4\u00e4ri\u00e4 n\u00e4ytteiss\u00e4 tutkittiin Western blotin avulla. Tuloksia analysoitiin Western blotin membraanikuvia hy\u00f6dynt\u00e4en. Seerumistimulaatio ja insuliinin kaltainen kasvutekij\u00e4 1 (eng. Insulin-like growth factor 1, IGF-1) lis\u00e4siv\u00e4t viljeltyjen ihmislihassolujen proteiinisynteesi\u00e4. Seerumistimulaatiossa lihassolujen medium sis\u00e4lsi enemm\u00e4n kasvutekij\u00f6it\u00e4 suhteessa kontrolliryhm\u00e4\u00e4n. Seerumistimulaatio lis\u00e4si 14C-glukoosin hiiliatomien kertymist\u00e4 proteiineihin ja RNA:han, kun taas insuliinin kaltainen kasvutekij\u00e4 1 lis\u00e4si 14C-glukoosin hiiliatomien kertymist\u00e4 RNA:han ja lipideihin. The stimulation of muscle size is a complex process, but understanding it is crucial for the development of treatment methods for patients suffering from muscle loss. In this Master\u2019s thesis, we investigated how to increase the protein synthesis of cultured human muscle cells and how glucose is utilized in the biomolecule synthesis of these muscle cells. We cultured human muscle cells and accomplished cell tests to increase the protein synthesis. We isolated proteins from the muscle cells and measured protein concentrations. Liquid scintillation counting was used to measure protein contents and study the incorporation of glucose into proteins, RNAs and lipids. We utilized Western blotting to discover possible differences in rates of phosphoglycerate dehydrogenase enzyme (PHGDH) and phosphorylated proteins S6K1 and S6. Phosphoglycerate dehydrogenase enzyme is an essential enzyme in serine synthesis pathway, and phosphorylated proteins S6K1 and S6 indicate the activation of the mechanistic target of rapamycin complex 1 (mTORC1). The results were analyzed using Western blotting membrane photos. Serum stimulation and insulin-like growth factor 1 (IGF-1) increased the protein synthesis of the muscle cells. In serum stimulation the medium of the muscle cells contained more growth factors compared to the medium of the control cells. Serum stimulation caused an increase in incorporation of 14C glucose carbon atoms into proteins and RNAs. Insulin-like growth factor 1 increased the incorporation of those carbon atoms into RNAs and lipids. Our results suggested that serum stimulation and insulin-like growth factor 1 may have potential to increase the growth of the cultured human muscle cells, which is an important result for further studies of treatment methods for muscle loss.
