Three-dimensional imaging (3D) using tissue clearing techniques aids in studying complex structures like vasculature. However, only one preliminary study has examined the 3D skeletal muscle vasculature using similar technique. Since this method has not been extensively studied before, this thesis aimed to develop the method for whole mouse muscles and to investigate the effects of exercise on skeletal muscle arterial network using 3D imaging in normal and overweight mice. Adult C57BL/6J male mice (n=40) were divided into four groups: lean sedentary, lean exercise, obese sedentary and obese exercise. Obesity, hyperglycemia and insulin resistance were induced using a Western diet for 18 weeks. The exercise groups had free access to running wheels for the last 9 weeks within their cages. The entire intervention lasted 18 weeks. Tibialis anterior muscles of mice (n=12, 3/group) were stained with SMA-Cy3 antibody to visualize the arterial network and cleared for 3D vascular structure analysis utilizing light sheet microscopy and Imaris software. Other methods utilized in this study included body composition analysis, glucose tolerance test and maximal running capacity test. Body weight and fat percentage were significantly increased in the obese mice. However, glucose tolerance was impaired only in the obese sedentary group (p<0.0001), demonstrating the metabolic health benefit of exercise in the obese mice. The development of the 3D imaging protocol was successful and visual analysis with Imaris enabled the exploration of the 3D arterial structure. The 3D quantitative analyses revealed no statistically significant differences in arterial characteristics such as area, volume, total length, branch length, diameter, branch level and depth between the groups. 3D visualization of muscles using whole mount staining and iDISCO tissue clearing protocol proved effective in analyzing skeletal muscle arterial vasculature. With this rather small sample size, the results indicate that the arterial network was not affected by exercise and obesity. Aikuiset C57BL/6J uroshiiret (n=40) jaettiin nelj\u00e4\u00e4n ryhm\u00e4\u00e4n: laihat liikkumattomat, laihat liikkuvat, ylipainoiset liikkumattomat ja ylipainoiset liikkuvat. Kokeen kokonaispituus oli 18 viikkoa. Lihavuus, hyperglykemia ja insuliiniresistenssi aiheutettiin koko kokeen ajan kest\u00e4v\u00e4ll\u00e4 korkearasvaisella ruokavaliolla. J\u00e4lkimm\u00e4iset 9 viikkoa liikkuvien ryhmill\u00e4 oli h\u00e4keiss\u00e4\u00e4n vapaa p\u00e4\u00e4sy juoksupy\u00f6rille. Hiirten etummaiset s\u00e4\u00e4rilihakset (n=12, 3/ryhm\u00e4) v\u00e4rj\u00e4ttiin SMA-Cy3-vasta-aineella ja tehtiin l\u00e4pin\u00e4kyv\u00e4ksi 3D verisuonirakenteen analysointia varten. 3D analyysit tehtiin valomikroskooppia ja Imaris-analyysiohjelmaa hy\u00f6dynt\u00e4en. Tutkimuksessa k\u00e4ytettiin my\u00f6s kehonkoostumusmittausta, glukoositoleranssitesti\u00e4 ja maksimaalista juoksutesti\u00e4 kuvaamaan hiirten fysiologisia ominaisuuksia. Ylipainoisilla hiirill\u00e4 kehon paino ja rasvaprosentti kasvoivat merkitt\u00e4v\u00e4sti. Kuitenkin glukoositoleranssi oli heikentynyt vain liikkumattomien ylipainoisten ryhm\u00e4ll\u00e4 (p<0,0001), mik\u00e4 osoittaa liikunnan positiivisen hy\u00f6dyn metabolisen terveyden kannalta ylipainoisilla hiirill\u00e4. 3D-kuvantamisprotokollan kehitys onnistui ja se mahdollisti visuaalisen ja numeraalisen 3D-valtimorakenteen analysoinnin Imaris-ohjelmaa hy\u00f6dynt\u00e4en. Kvantitatiiviset analyysit eiv\u00e4t paljastaneet tilastollisesti merkitsevi\u00e4 eroja valtimoiden pinta-alassa, tilavuudessa, kokonaispituudessa, haaran pituudessa, halkaisijassa, haaran tasoilla eik\u00e4 haarojen syvyydess\u00e4. Lihasten 3D-visualisointi koko kudoksen v\u00e4rj\u00e4yksell\u00e4 ja iDISCO-kudosl\u00e4p\u00e4isytekniikalla osoittautui tehokkaaksi protokollaksi luurankolihaksen verisuoniston analysoinnissa.
