It is considered a lifestyle disease. Colorectal cancer is divided into right-sided and left-sided cancer tumours depending on their location in the colon. Most tumours are left-sided, but the right-sided ones have a worse prognosis. Side differences include gut microbiota and inflammation levels. Right-sided tumours have a higher inflammation rate and they have been shown to contain more bacteria-related biofilms. It has been demonstrated that biofilm increases cytokines Interleukin-6 and CCL2/MCP-1 in the tumour microenvironment, which are the most common cause of inflammation in colorectal cancer. The reasons for tumour sidedness are largely unclear, which is why the effects of sidedness on microbiota and inflammatory factors in colorectal cancer were studied in this thesis. The samples and data were collected from eight colorectal cancer patients who were operated on in the Central Finland Health Care District. They were voluntarily recruited to SUOPA research of which this MSc thesis is part. From stool and tumour samples the microbiota was analysed by sequencing the bacterial 16S rRNA gene. The gene expression of cytokines and Toll-like receptor 5 from tumour samples was defined by quantitative real-time PCR. Statistical analyses of gene expression were done with the SPSS program and statistical analyses of the microbiota with the CLC Microbial Genomics program. The small sample size proved to be challenging, not providing significance in gene expression. However, statistical significance was observed in one bacterial order, Clostridiales (p=0,02), and in five bacterial genera including Family XIII AD3011 group (p=0,05), Roseburia (p=0,02), Ruminiclostridium 9 (p=0,03), four Asteroleplasma (p=0,02; p=0,03; p=0,04 & p=0,05) and two Christensenellaceae R-7 group (p=0,03 & p=0,04) of OTUs. Most of these belong to Firmicutes, which is known to be elevated in patients with colorectal cancer. All the bacteria have been associated with increasing levels in left-sided tumour patients, but the role of some of them in gut and colorectal cancer is not well studied. Suurin osa kasvaimista on vasemmanpuoleisia, mutta oikeanpuoleisen kasvaimen potilailla on huonompi ennuste. Kasvaimissa on havaittu puoleisuuseroja muun muassa mikrobiston koostumuksessa ja tulehdustilassa. Oikeanpuoleisiin kasvaimiin liittyy useammin my\u00f6s vakavia tulehdustiloja ja niiss\u00e4 on havaittu runsasta mikrobikerrostuman eli biofilmin muodostusta. Biofilmin tiedet\u00e4\u00e4n lis\u00e4\u00e4v\u00e4n sytokiinien CCL2/MCP-1 ja Interleukiini-6:n m\u00e4\u00e4r\u00e4\u00e4. Tarkkoja syit\u00e4 kasvainten puoleisuuseroille ei kuitenkaan tiedet\u00e4, joten t\u00e4ss\u00e4 tutkielmassa tutkittiin edelleen puoleisuuseroja suoliston mikrobikoostumuksessa ja tulehdustekij\u00f6iden ilmentymisess\u00e4. Aineisto ker\u00e4ttiin kahdeksalta Keski-Suomen sairaanhoitopiirin kirurgian yksil\u00f6ss\u00e4 leikattavilta potilailta, jotka vapaaehtoisesti rekrytoitiin SUOPA-tutkimukseen, jota Pro gradu -tutkielma on osana. Potilaiden ulosten\u00e4ytteist\u00e4 ja kasvaimista analysoitiin mikrobikoostumus sekvensoimalla bakteerien \n16S rRNA -geeni. Sytokiinien ja Tollin-kaltaisen reseptori 5:n geeniekspressio m\u00e4\u00e4ritettiin kasvainn\u00e4ytteist\u00e4 kvantitatiivisella PCR:ll\u00e4. Tilastolliset analyysit suoritettiin geeniekspression osalta SPSS-ohjelmistolla ja mikrobikoostumuksen osalta CLC Microbial Genomics -ohjelmistopaketilla. Pieni otoskoko osoittautui tilastollisesti haastavaksi, joten geeniekspression suhteen puoleisuuseroissa ei havaittu merkitsev\u00e4\u00e4 ero. Merkitt\u00e4v\u00e4 ero havaittiin kasvainn\u00e4ytteiss\u00e4 yhden bakteerilahkon, Clostridiales (p=0,02), sek\u00e4 viiden bakteerisuvun; Family XIII AD3011 group (p=0,05), Roseburia (p=0,02), Ruminiclostridium 9 (p=0,03) sek\u00e4 nelj\u00e4n Asteroleplasma (p=0,02; p=0,03; p=0,04 & p=0,05) ja kahden Christensenellaceae R-7 group (p=0,03 & p=0,04) -suvulle kuuluvan OTU:n suhteen. Suurin osa bakteereista kuuluu firmikuuttien p\u00e4\u00e4jaksoon, jonka m\u00e4\u00e4r\u00e4n tiedet\u00e4\u00e4n kohonneen paksusuolensy\u00f6v\u00e4n potilailla. Merkitt\u00e4v\u00e4t bakteerit esiintyv\u00e4t oikeanpuoleisen kasvaimen potilailla runsaimpina, mutta tutkimuksissa ne linkittyv\u00e4t suurempaan esiintyvyyteen vasenpuoleisen kasvaimen potilailla, tosin kaikkien roolia suolistossa ja paksusuolensy\u00f6v\u00e4ss\u00e4 ei ole yht\u00e4 hyvin tutkittu.
