Menopaussin aikaisella liikuntaharjoittelulla on todettu olevan positiivinen vaikutus terveysvaivoihin, mutta yksin se ei riit\u00e4 est\u00e4m\u00e4\u00e4n terveydess\u00e4 tapahtuvia haitallisia muutoksia. Siksi on t\u00e4rke\u00e4\u00e4 tutkia, muuttuuko systeeminen vaste liikunnalle menopaussin yhteydess\u00e4, sek\u00e4 millaisia vaikutuksia n\u00e4ill\u00e4 muutoksilla on kudostasolla. Viestin v\u00e4litt\u00e4jin\u00e4 solunulkoisilla vesikkeleill\u00e4 (EV) on t\u00e4rke\u00e4 rooli, kun taas mikro-RNA:t (miR) ovat merkitt\u00e4vi\u00e4 geenien s\u00e4\u00e4telyss\u00e4. Yhdess\u00e4 ne ovat sek\u00e4 olennainen osa kudosten v\u00e4lisess\u00e4 vuorovaikutuksessa ett\u00e4 sopeutumisessa harjoitteluun. T\u00e4m\u00e4n tutkimuksen tarkoitus on selvitt\u00e4\u00e4 ensinn\u00e4kin sit\u00e4, vaikuttaako naarasrottien munasarjojen poistolla aikaansaatu munasarjahormonien puute akuuttiin liikuntavasteeseen ja toiseksi sit\u00e4, havaitaanko muutoksia kudostasolla maksan proteiinien ilmentymisess\u00e4. Tutkimuksessa eristettiin plasmasta EV:t k\u00e4ytt\u00e4en ultrasentrifugointia ja kokoerottelukromatografiaa, joista eristettiin RNA, sek\u00e4 tehtiin miR kirjastot. Kirjastot sekvensoitiin ja tuloksia vertailtiin etsien eroja EV:iden miR kuormissa ryhmien v\u00e4lill\u00e4. Lis\u00e4ksi maksan kudosn\u00e4ytteist\u00e4 tehtiin Western Blot proteiinien ilmentymistasojen muutosten selvitt\u00e4miseksi. Tutkimuksessa havaittiin muutoksia kahden ryhm\u00e4vertailun miR kuormassa. Ensimm\u00e4inen vertailu kertoi, vaikuttiko munasarjojen poisto miR signalointiin, jossa yhdeks\u00e4n miR:n ilmentymistaso oli muuttunut merkitt\u00e4v\u00e4sti. N\u00e4ist\u00e4 let-7g-5p, miR-100-5p ja -145-5p havaittiin olevan yhteydess\u00e4 tyypin 2 diabeteksen signalointireittiin. Toisessa vertailussa selvitettiin, oliko akuutilla liikuntasuorituksella vaikutusta miR:iden signalointiin. T\u00e4ss\u00e4 vertailussa havaittiin, kolmen miR:n merkitt\u00e4v\u00e4sti muuttunut ilmentymistaso. N\u00e4ist\u00e4 miR-100-5p todettiin olevan yhteydess\u00e4 MAPK signalointireittiin, jonka tiedet\u00e4\u00e4n liittyv\u00e4n harjoitteluun. Tarkastellessa akuutin liikuntasuorituksen vaikutusta miR signalointiin munasarjojen poiston j\u00e4lkeen ryhmien v\u00e4lill\u00e4 ei n\u00e4hty muutosta miR:issa. My\u00f6sk\u00e4\u00e4n maksan proteiinien ilmentymisess\u00e4 ei l\u00f6ydetty merkitsevi\u00e4 muutoksia. Hence, it is important to study whether systemic response to exercise changes during menopause and if the changes have an impact at the tissue level. Extracellular vesicles (EVs) are important signal carriers, while microRNAs (miRs) have a significant role in regulating gene expression. These two elements are key factors in mediating communication between tissues and are essential for the adaptation process during exercise. This study aims to investigate whether the decrease in ovarian hormones resulting from ovariectomy (OVX) impacts the acute exercise response in female rats and if these changes can be detected in the protein expression of the liver. In this study, EVs were isolated from rat plasma through ultracentrifugation and size-exclusion chromatography. Subsequently, RNA was extracted, and miR libraries were constructed. The libraries were sequenced, and the results were compared to identify differences via DESeq2 in miR load between groups. The target proteins of the differentially expressed miRs were explored using the MirPath v.3 analysis tool. Also, Western Blot analysis was conducted on liver tissue samples to determine whether miRs affected the protein expression of their target proteins. We observed changes in miR expression in two group comparisons. The first comparison addressed the question of whether OVX affects miR signaling. The expression of nine miRs changed significantly, and miRs let-7g-5p, miR-100-5p, and -145-5p were found to be linked to the type 2 diabetes signaling pathway. The second comparison revealed the impact of acute exercise on miR signaling, resulting in a significant change in the expression of three miRs in the sham group. Among these, miR-100-5p was identified to be associated with the MAPK signaling pathway, which is related to exercise. No changes in miR expression were observed when examining the impact of acute exercise on miR signaling after OVX. Additionally, no significant changes were found in the protein expression of the liver.
