Viestin välittäjinä solunulkoisilla vesikkeleillä (EV) on tärkeä rooli, kun taas mikro-RNA:t (miR) ovat merkittäviä geenien säätelyssä. Yhdessä ne ovat sekä olennainen osa kudosten välisessä vuorovaikutuksessa että sopeutumisessa harjoitteluun. Tämän tutkimuksen tarkoitus on selvittää ensinnäkin sitä, vaikuttaako naarasrottien munasarjojen poistolla aikaansaatu munasarjahormonien puute akuuttiin liikuntavasteeseen ja toiseksi sitä, havaitaanko muutoksia kudostasolla maksan proteiinien ilmentymisessä. Tutkimuksessa eristettiin plasmasta EV:t käyttäen ultrasentrifugointia ja kokoerottelukromatografiaa, joista eristettiin RNA, sekä tehtiin miR kirjastot. Kirjastot sekvensoitiin ja tuloksia vertailtiin etsien eroja EV:iden miR kuormissa ryhmien välillä. Lisäksi maksan kudosnäytteistä tehtiin Western Blot proteiinien ilmentymistasojen muutosten selvittämiseksi. Tutkimuksessa havaittiin muutoksia kahden ryhmävertailun miR kuormassa. Ensimmäinen vertailu kertoi, vaikuttiko munasarjojen poisto miR signalointiin, jossa yhdeksän miR:n ilmentymistaso oli muuttunut merkittävästi. Näistä let-7g-5p, miR-100-5p ja -145-5p havaittiin olevan yhteydessä tyypin 2 diabeteksen signalointireittiin. Toisessa vertailussa selvitettiin, oliko akuutilla liikuntasuorituksella vaikutusta miR:iden signalointiin. Tässä vertailussa havaittiin, kolmen miR:n merkittävästi muuttunut ilmentymistaso. Näistä miR-100-5p todettiin olevan yhteydessä MAPK signalointireittiin, jonka tiedetään liittyvän harjoitteluun. Tarkastellessa akuutin liikuntasuorituksen vaikutusta miR signalointiin munasarjojen poiston jälkeen ryhmien välillä ei nähty muutosta miR:issa. Myöskään maksan proteiinien ilmentymisessä ei löydetty merkitseviä muutoksia. Tulosten perusteella voidaan päätellä, että estrogeenin puute estää naarasrotilla akuutin liikunnan aiheuttaman vasteen miR signaloinnissa, joka tukee aiemmin havaittua löydöstä naisilla. Menopause is associated with diverse negative effects on women’s health, and while exercise can mitigate some of these changes, it cannot prevent all the unfavorable impacts on health. Hence, it is important to study whether systemic response to exercise changes during menopause and if the changes have an impact at the tissue level. Extracellular vesicles (EVs) are important signal carriers, while microRNAs (miRs) have a significant role in regulating gene expression. These two elements are key factors in mediating communication between tissues and are essential for the adaptation process during exercise. This study aims to investigate whether the decrease in ovarian hormones resulting from ovariectomy (OVX) impacts the acute exercise response in female rats and if these changes can be detected in the protein expression of the liver. In this study, EVs were isolated from rat plasma through ultracentrifugation and size-exclusion chromatography. Subsequently, RNA was extracted, and miR libraries were constructed. The libraries were sequenced, and the results were compared to identify differences via DESeq2 in miR load between groups. The target proteins of the differentially expressed miRs were explored using the MirPath v.3 analysis tool. Also, Western Blot analysis was conducted on liver tissue samples to determine whether miRs affected the protein expression of their target proteins. We observed changes in miR expression in two group comparisons. The first comparison addressed the question of whether OVX affects miR signaling. The expression of nine miRs changed significantly, and miRs let-7g-5p, miR-100-5p, and -145-5p were found to be linked to the type 2 diabetes signaling pathway. The second comparison revealed the impact of acute exercise on miR signaling, resulting in a significant change in the expression of three miRs in the sham group. Among these, miR-100-5p was identified to be associated with the MAPK signaling pathway, which is related to exercise. No changes in miR expression were observed when examining the impact of acute exercise on miR signaling after OVX. Additionally, no significant changes were found in the protein expression of the liver. We conclude that estrogen deficiency blunts the acute exercise-induced response in miR signaling in female rats and supports previous findings observed in women.
