Koska j\u00e4nteiden solut k\u00e4ytt\u00e4v\u00e4t happea ja sis\u00e4lt\u00e4v\u00e4t energia-aineenvaihduntaj\u00e4rjestelm\u00e4n entsyymiketjuja, niiden uskotaan k\u00e4ytt\u00e4v\u00e4n glukoosia energia-aineenvaihdunnassaan. Positroniemissiotomografia (PET) on radioaktiivisia isotooppeja hy\u00f6dynt\u00e4v\u00e4 kuvantamismenetelm\u00e4, jolla voidaan tutkia elimist\u00f6n biokemiallisiaja aineenvaihdunnallisia tapahtumia in vivo. T\u00e4m\u00e4n tutkimuksen tarkoituksen on positroniemissiotomografian ja [18F]FDG:n avulla tutkia akillesj\u00e4nteen glukoosin k\u00e4ytt\u00f6\u00e4 eri rasitustasoilla ja verrata sit\u00e4 luustolihaksen glukoosin k\u00e4ytt\u00f6\u00f6n. Tutkimukseen osallistui kahdeksan vapaaehtoista miest\u00e4 (ik\u00e4 29 \u00b1 7 v.; V02max 51 \u00b1 9 ml*kg-1min). Kaikki koehenkil\u00f6t tutkittiin kolmena eri p\u00e4iv\u00e4n\u00e4 v\u00e4hint\u00e4\u00e4n kahden p\u00e4iv\u00e4n v\u00e4lein kerran kullakin kolmesta rasitustasosta 30 % (LOW), 55 % (MID) ja 75 % (HIGH) V02max:sta. PET-tutkimus alkoi kymmenen minuuttia polkupy\u00f6r\u00e4rasituksen alkamisesta, jolloin koehenkil\u00f6n kyyn\u00e4rlaskimoon injisoitiin [18F]FDG -merkkiainetta. [18F]FDG:n injisoinnin j\u00e4lkeen polkupy\u00f6r\u00e4rasitusta jatkettiin viel\u00e4 25 minuuttia. Polkupy\u00f6r\u00e4rasituksen j\u00e4lkeen mitattiin akillesj\u00e4nteen, nelip\u00e4isen reisilihaksen sek\u00e4 lyhyen pohjeluulihaksen glukoosin k\u00e4ytt\u00f6 PET:n avulla. Koehenkil\u00f6iden syke ja laktaattikonsentraatio kasvoivat merkitt\u00e4v\u00e4sti eri rasitustasojen v\u00e4lill\u00e4 (p < 0.001). Plasman glukoosikonsentraatiossa ei havaittu eroja eri mittauskertojen eik\u00e4 eri rasitustasojen v\u00e4lill\u00e4. FDG kertyi lihaksiin ja akillesj\u00e4nteeseen ja sen kulutus voitiin laskea graafisen analyysin avulla. Akillesj\u00e4nteen glukoosin kulutus oli 7.1 \u00b1 1.5, 6.6 \u00b1 1.1 ja 6.0 \u00b1 1.1 umol*kg-lmin-1 ja lyhyen pohjeluulihaksen 13.3 \u00b1 5.4, 11.4 \u00b1 5.4 ja 11.5 \u00b1 2.9 umol*kg-1min-l eri rasitustasoilla (LOW, MID ja HIGH). Akillesj\u00e4nteen ja lyhyen pohjeluulihaksen glukoosin k\u00e4yt\u00f6ss\u00e4 ei havaittu muutoksia eri rasitustasojen v\u00e4lill\u00e4. Lyhyen pohjeluulihaksen glukoosin k\u00e4ytt\u00f6 oli kuitenkin akillesj\u00e4nteen glukoosin k\u00e4ytt\u00f6\u00e4 suurempaa LOW - (p < 0.05) ja HIGH-tasoilla (p < 0.05). Nelip\u00e4isen reisilihaksen glukoosin k\u00e4ytt\u00f6 kasvoi rasituksen intensiteetin my\u00f6t\u00e4 47.9 \u00b1 34.8, 120.1 \u00b1 34.6 ja 152.3 \u00b1 74.0 umol*kg-lmin-l. Nelip\u00e4isen reisilihaksen glukoosin k\u00e4ytt\u00f6 kasvoi LOW -tasolta MID-tasolle l\u00e4hes kolminkertaisesti ja LOW-tasolta HIGH-tasolle yli nelinkertaisesti, molemmissa (p < 0.001). Nelip\u00e4isen reisilihaksen glukoosin k\u00e4ytt\u00f6 oli merkitt\u00e4v\u00e4sti suurempaa (p < 0.001) verrattuna akillesj\u00e4nteen tai lyhyen pohjeluulihaksen glukoosin k\u00e4ytt\u00f6\u00f6n kaikilla kolmella rasitustasolla. T\u00e4m\u00e4n tutkimuksen tulosten perusteella akillesj\u00e4nne k\u00e4ytt\u00e4\u00e4 glukoosia energiakseen ja akillesj\u00e4nteen glukoosin k\u00e4ytt\u00f6 voidaan mitata PET:n avulla. Kuormituksessa aktiivisen luustolihaksen glukoosin k\u00e4ytt\u00f6 kasvaa rasituksen intensiteetin lis\u00e4\u00e4ntyess\u00e4.
