Suurin osa tutkimuksesta on painottunut iäkkäisiin, henkilöihin, joilla on ylipainoa ja urheilijoihin sekä korkeaan liikuntamäärään. Laajalla väestöllä tehdyt tutkimukset ovat olleet vähäisiä. Tämän poikkileikkaustutkimuksen tavoitteena oli tutkia vapaa-ajan liikunnan ja suolistomikrobiston välistä yhteyttä. Suolistomikrobistodata ulostenäytteistä ja vapaa-ajan liikunta-aktiivisuus kerättiin 302 henkilöltä, jotka osallistuivat Sepelvaltimotaudin riskitekijöiden interventioprojektin (STRIP) seurantaan 26-vuotiaina. Tiedot vapaa-ajan liikunnasta kerättiin itsetäytettävällä kyselyllä. Suolistomikrobinäytteet profiloitiin käyttämällä 16S rRNA sekvenssiteknologiaa. Kaikki tutkimuksen analyysit suoritettiin R ohjelmassa soveltuvilla mikrobianalyysipaketteja. Vapaa-ajan liikunta-aktiivisuuden perusteella henkilöt jaettiin seuraaviin ryhmiin: matalan liikunta-aktiivisuuden ryhmä (n = 83, ≤5MET/h/vko) ja korkean liikunta-aktiivisuuden ryhmä (n = 219,≥16MET/h/vko) Alfadiversiteettiä tutkittiin Shannonin indeksillä sekä Wilcoxonin testillä. Betadiversiteetin analyysi tehtiin permutaatio varianssianalyysillä (PERMANOVA), käyttäen adonis2 funktiota vegan-paketissa, sekä Bray-Curtisin ja Jaccardin dissimilariteetti-indeksejä. Differentiaalista esiintymistiheyttä tutkittiin käyttämällä DESeq2 ja ALDEx2 menetelmiä. Alfa- ja betadiversiteetti analyysit suoritettiin koko datalle. DESeq2 ja ALDEx2 analyysit tehtiin vähäisen pitoisuuden lajit poistettuina analyysista. Tulosten visualisointi tehtiin R-ohjelmassa moniulotteisella skaalauksella. Raportoinnissa hyödynnettiin soveltuvin osin STORM-ohjeistusta. Havaitsimme, että vaikka alfadiversiteetti oli ryhmien välillä samanlainen, betadiversiteetti erosi merkitsevästi. Eroa betadiversiteetissä ajoivat Prevotella ja Paraprevotella korkean, sekä Bacteroides matalan liikunta-aktiivisuuden ryhmässä. Esiintymistiheysanalyysissa DESeq2 tunnisti 18 ja ALDEx2 kolme sukua, jotka erosivat merkitsevästi ryhmien välillä. Kohtalainen liikunta-annos on yhteydessä betadiversiteettiin ja esiintymistiheyden lisääntymiseen tietyissä suvuissa, kuten Prevotella-, Veillonella- ja Lactobacillus, jotka on aikaisemmin yhdistetty kuormittavampaan liikuntaan. There is increasing evidence of interaction between physical activity and gut microbiota. Research data suggest of beneficial effects of physical activity to gut microbiota and potential effect of gut microbiota to e.g., physical performance. However, the majority of the current research has focused on elderly, people who are overweight and athletes, whereas general population research is scarce. The aim of this cross-sectional study was to explore the association between leisure time physical activity (LTPA) and gut microbiota (GM) composition.   Gut microbiota from stool samples and LTPA were assessed in 302 adults aged 26 years who participated in the longitudinal Special Turku Coronary Risk Factor Intervention Project (STRIP). LTPA was assessed with a self-administrated questionnaire. Profiling of gut microbiota was done using 16S rRNA amplicon sequencing.  All statistical analyses were performed with R programming language using suitable microbiome analysis packages. Based on their LTPA participants were divided in low (n = 83, ≤5MET/h/wk) and high (n = 219, ≥16MET/h/wk) PA groups. Alpha diversity was assessed by counting abundances using Shannon Index in microbiome and further Wilcoxon test. Beta diversity was analysed with Permutational Analysis of Variation (PERMANOVA) using adonis2 function in vegan and Bray-Curtis and Jaccard dissimilarity indexes. Beta dispersion analysis was done using PERMDISP2 in vegan package. For differential abundance analyses DESeq2 and ALDEx2 were used. The alpha and beta diversity analyses were performed to all data, including rare taxa. DeSeq2 and ALDEx2 were performed with rare taxa removed. Data was visualised in R with Multidimensional Scaling. Reporting occurred according to STORM guidelines as applicable. We found that even though alpha diversity was similar between the high and low activity groups, beta diversity differs significantly between high and low activity groups. The difference was driven with Prevotella and Paraprevotella species in the high and genus Bacteroides in the low PA group. In differential abundance analysis DESeq2 identified 18 ja ALDEx2 three generas that were significantly different between the PA groups. A moderate dosage of LTPA is associated with beta diversity and increased abundance of certain genera, such as Prevotella-, Veillonella- and Lactobacillus, that are previously associated with more vigorous PA.
