Hyönteisten levinneisyyden ja selviytymisen kannalta elinympäristön lämpötila on ehkä tärkein abioottinen tekijä, ja varsinkin matalat lämpötilat ovat haasteellisia alueilla, joilla ilmastolliset vaihtelut ovat suuria. Tässä projektissa selvitettiin Trypanosomatidae-loisten vaikutuksia stressinkestävän Drosophila lummei - mahlakärpäsen kylmänkestävyyteen, liikeaktiivisuuteen ja hengissä selviytymiseen. Aihetta tutkittiin geneettisillä (kvantitatiivinen PCR) ja fenotyyppisillä (liikeaktiivisuus- ja selviytymiskokeet) testeillä. Kylmänkestävyys ei korreloinut infektion määrän kanssa kummallakaan sukupuolella, eikä kylmäkäsittelyä edeltävällä ja sitä parantavalla akklimaatiokäsittelyllä ollut vaikutusta loisinfektion määrään. Infektion määrä sen sijaan oli matalampi naarailla kuin koirailla. Liikeaktivisuus oli suurempaa ei-infektoituneilla kärpäsillä verrattuna infektoituneisiin, mutta tutkimuksissa käytetty valojakso ei vaikuttanut kärpästen aktiivisuuteen. Selviytymiskokeessa infektoitujen kärpästen elinikä oli lyhyempi kuin ei-infektoitujen, mutta valojakso ei vaikuttanut elinikään. Saadaksemme uutta tietoa hyönteisten immuunipuolustuksesta, tarvitaan myös alkueläininfektiotutkimuksia koska suuri osa aiemmista tutkimuksista on keskittynyt bakteereihin ja viruksiin Trypanosomatids are an extremely diverse group of single cell eukaryotic parasites infecting humans, animals and plants. Insect trypanosomatids are a broad but a less well-known group of monoxeus parasites, that affect a range of their hosts’ fitness traits, including feeding behavior and immunity. Also less is known about the effects of these infections on host's ability to respond to abiotic factors. Temperature is perhaps the most important environmental factor governing insect distributions, fitness and survival and particularly low temperatures appear challenging for insect physiology in habitats with drastic seasonal change. The aim of this study was to investigate the effect of trypanosomatid infection on cold tolerance, survival and locomotor activity of the flies from highly stress tolerant Drosophila virilis group species, Drosophila lummei. Genetic (quantitative PCR) and phenotypic methods (locomotor activity and survival experiments) were used to study these questions. Cold tolerance was not affected by infection load in neither sex nor different acclimation treatment groups, while females had lower infection load compared to males. Locomotor activity results indicated that there was a difference in total activity between infected and non-infected flies, but light cycle did not influence total activity. Moreover, infected flies had shorter lifespan than non-infected flies, but again light cycle did not affect the survival. Studying gut infecting trypanosomatids in Drosophila is important as most Drosophila infection studies are only performed with bacteria and virus. Also, as trypanosomatids are known to be common parasites in insects, studies of their effects on their hosts could reveal new interesting aspects of insect immune system.
