ISO 6579-1 standardin mukainen bakteeriviljely on ollut toistaiseksi ainoa menetelm\u00e4 bakteerin tunnistukseen alkutuotannon n\u00e4ytematriiseista, mutta huhtikuussa 2021 voimaan tullut asetus zoonooseista (316/2021) mahdollistaa valvontaan kuuluvien n\u00e4ytteiden tutkimisen my\u00f6s muilla laboratoriomenetelmill\u00e4. Nopea tunnistus on t\u00e4rke\u00e4\u00e4, jotta bakteerin laaja levi\u00e4minen voidaan toimenpiteill\u00e4 varhaisessa vaiheessa ehk\u00e4ist\u00e4. Toimenpiteist\u00e4 huolimatta bakteeri voi s\u00e4ily\u00e4 elinkykyisen\u00e4 tiloilla pitk\u00e4\u00e4n, jota voi selitt\u00e4\u00e4 bakteerien kyky muodostaa biofilmi\u00e4. T\u00e4ss\u00e4 tutkielmassa vertailtiin ISO 6579-1 standardin mukaisen bakteeriviljelyn ja kaupallisen qPCR-menetelm\u00e4n soveltuvuutta Salmonella-bakteerin osoitukseen alkutuotannon n\u00e4ytematriiseilla. Lis\u00e4ksi tutkittiin tuotantotiloilta eristettyjen Salmonella-kantojen biofilmin muodostusta kristalliviolettiv\u00e4rj\u00e4ysmenetelm\u00e4ll\u00e4 kahdessa eri l\u00e4mp\u00f6tilassa sek\u00e4 kolmessa eri kasvatusliuoksen pitoisuudessa. Menetelm\u00e4vertailun n\u00e4yteotantaan kuului pakastettuja sek\u00e4 tuoreita naudan uloste- ja ymp\u00e4rist\u00f6n\u00e4ytteit\u00e4. Menetelmien havaitsemisraja (LOD) m\u00e4\u00e4ritettiin kanan ja naudan ulosten\u00e4ytteill\u00e4 sek\u00e4 p\u00f6lyn\u00e4ytteill\u00e4. N\u00e4ytteiden esirikastus tehtiin kahdella rikastusliemell\u00e4, joiden vaikutusta menetelmien herkkyyteen arvioitiin. Menetelm\u00e4vertailun ja havaitsemisrajan m\u00e4\u00e4rityksen perusteella qPCR-menetelm\u00e4 soveltui yht\u00e4 hyvin kuin bakteeriviljely Salmonella-bakteerin osoitukseen tuoreista naudan uloste- ja ymp\u00e4rist\u00f6n\u00e4ytteist\u00e4. Bakteeriviljely sen sijaan soveltui luotettavammin bakteerin osoitukseen kanan ulosten\u00e4ytteist\u00e4. Kummallakaan rikastusliemell\u00e4 ei ollut vaikutusta menetelmien herkkyyteen. qPCR-menetelm\u00e4 mahdollistaa tuloksen valmistumisen kahden vuorokauden sis\u00e4ll\u00e4, kun vastaavasti viljelyll\u00e4 tuloksen valmistumiseen kuluu 3-6 vuorokautta. Biofilmin muodostuksen tutkiminen osoitti, ett\u00e4 l\u00e4hes kaikki tutkitut kannat muodostivat biofilmi\u00e4 huoneenl\u00e4mm\u00f6ss\u00e4 ja alhaisessa ravinneliuoksen pitoisuudessa. The ISO 6579-1 standard bacterial culture has been so far the only method to detect Salmonella from primary production samples, but the decree on zoonoses (316/2021), which entered into force in April 2021, allows other laboratory methods to be used as well. Rapid diagnostic methods are needed to prevent the spread of the bacterium. Salmonella can persist long-term in production farms despite the preventing procedures and biofilm formation may facilitate the long-term persistence. This thesis compared the suitability of ISO 6579-1 bacterial culture and the real-time PCR method to detect Salmonella from primary production sample matrices. In addition, the ability of Salmonella strains to form biofilm was studied with crystal violet assay. The biofilm formation was assessed for strains isolated from Finnish farms in two different temperatures and three different concentration mediums. The comparative study of the methods included frozen and fresh cattle feces and environmental samples. The limit of detection (LOD) for both methods were determined as well with cattle and poultry feces and dust samples. Two different enrichment broths were used for sample enrichment to evaluate their possible effect on sensitivity. Based on the method comparison and the limit of detection, the qPCR method was equally suitable and sensitive as the bacterial culture for Salmonella detection from fresh cattle feces and environmental samples. However, the bacterial culture was more suitable for the detection of the bacterium from poultry fecal samples. Neither enrichment broth had an effect on the sensitivity of the methods. The qPCR method enables faster results within two days while with bacterial culture the results are obtained within 3-6 days. The biofilm assay indicated that almost all strains could form biofilm at room temperature, and in a low nutrient concentration medium.
