Rapuja pyydet\u00e4\u00e4n useimmiten erityyppisill\u00e4 sy\u00f6titett\u00e4vill\u00e4 merroilla. T\u00e4ss\u00e4 tutkimuksessa arvioidaan joki- ja t\u00e4pl\u00e4rapujen esiintymisen havainnointia perinteisten rapumertojen lis\u00e4ksi uusilla ilman sy\u00f6tti\u00e4 pyyt\u00e4vill\u00e4 kolopyydyksill\u00e4, visuaalisella havainnoinnilla sek\u00e4 ymp\u00e4rist\u00f6-DNA-menetelm\u00e4n (eDNA) avulla. eDNA-menetelm\u00e4 on hy\u00f6dyllinen erityisesti uhanalaisten ja hankalasti havainnoitavien lajien tutkimuksessa, sill\u00e4 sen avulla on mahdollisuus saada tietoa ilman kohdelajin pyydyst\u00e4mist\u00e4. Perinteisi\u00e4 rapumertoja pidettiin pyynniss\u00e4 24 tuntia kullakin kohteella, kun taas kolopyydykset olivat pyynniss\u00e4 2 viikkoa, jotta ravut ehtiv\u00e4t tottua niihin. Visuaalinen havainnointi suoritettiin kahlaamalla tai tarkastelemalla veneest\u00e4 rapuja silm\u00e4m\u00e4\u00e4r\u00e4isesti ja vesikiikarin avulla. eDNA-n\u00e4ytteenotto suoritettiin siihen suunnitellulla j\u00e4rjestelm\u00e4ll\u00e4 pumppaamalla 5 L vett\u00e4 suodattimen l\u00e4pi. eDNA-analysointi tehtiin laboratoriossa qPCR-analyysin avulla. Lis\u00e4ksi kohdevesist\u00e4 otettiin vesin\u00e4ytteit\u00e4 tukemaan tuloksia. Perinteisill\u00e4 rapumerroilla ja eDNA-menetelm\u00e4ll\u00e4 saatiin suurimmilta osin yhtenevi\u00e4 tuloksia ja n\u00e4m\u00e4 menetelm\u00e4t todettiin toimiviksi havainnointimenetelmiksi. eDNA:n todettiin olevan kehityskelpoinen menetelm\u00e4 kertomaan, onko vesist\u00f6ss\u00e4 tietty\u00e4 rapulajia. Kolopyydykset eiv\u00e4t toimineet toivotulla tavalla, sill\u00e4 niihin meni vain kahdessa paikassa vain yksitt\u00e4isi\u00e4 rapuja. Visuaalinen havainnointi osoittautui huonoksi havainnointimenetelm\u00e4ksi etenkin syvill\u00e4 ja tummavetisill\u00e4 kohteilla. Kumpikaan n\u00e4ist\u00e4 menetelmist\u00e4 ei siis osoittautunut luotettavaksi havainnointimenetelm\u00e4ksi.", "language": "fi", "element": "description", "qualifier": "abstract", "schema": "dc"}, {"key": "dc.description.abstract", "value": "In Finland, three species of crayfish live in freshwater: noble crayfish (Astacus astacus), introduced signal crayfish (Pacifastacus leniusculus) and narrow-clawed crayfish (Pontastacus leptodactylus). Crayfish are most often caught with different types of traps. This study evaluates the detection of noble and signal crayfish with new non-baited tube traps, visual detection, and environmental-DNA (eDNA) in addition to traditional traps in lakes and rivers. The eDNA method is particularly useful in the study of endangered and difficult-to-detect species, as it provides access to information without capturing the target species. Traditional traps were kept for 24 hours in each watersystem, while tube traps were kept for 2 weeks in order to allow the crayfish to get used to them. Visual observation was performed by wading or viewing crayfish from the boat visually and using a water binocular. The eDNA sampling was performed with a system designed for it, by pumping 5 L of water through a filter. The eDNA analysis was performed in the laboratory using qPCR-analysis. In addition, water samples were taken from the target waters to support the results. Traditional traps and the eDNA-method yielded largely consistent results and were found to be viable detection methods. eDNA was identified as a viable method that should be encouraged. The tube traps did not work as desired, as only in two places only few crayfish went into them. Visual observation proved to be a poor method of observation as well, especially in deep and dark water objects.
