Acinetobacter baumannii on Maailman terveysjärjestön prioriteettipatogeeni. Se on herkästi antibiooteille resistentiksi tuleva bakteeri, joka aiheuttaa vakavia opportunistisia infektioita. Tässä kandidaatintutkielmassa selvitettiin faagiresistenssin muodostumisen vaikutuksia Acinetobacter baumannii -sairaalabakteerin kasvuominaisuuksiin. Potilasnäytteistä eristetyt Acinetobacter baumannii -kannat altistettiin niitä infektoiville faageille. Resistenssin kehittäneiden ja altistamattomien kantojen kasvuominaisuuksia vertailtiin spektrofotometrisesti Bioscreen C -mittauksella. Resistenssin kehittymisen havaittiin pääosin heikentävän bakteerien kasvuominaisuuksia. Suurin saavutettu bakteerimäärä pieneni jokaisella resistentillä kannalla ja suurin kasvunopeus heikentyi lähes jokaisella resistentillä kannalla verrattuna altistamattomiin kantoihin. Kasvun heikentyminen on yhteydessä muun muassa bakteerin virulenssin alenemiseen ja antibioottiherkkyyden kasvuun. Tutkimuksesta saatuja tuloksia voidaan näin ollen hyödyntää faagiterapian ja antibioottien yhdistelmähoitojen kehittämisessä. Antibiotic resistance is a growing concern among treatment of infections caused by bacteria. Phages have been proposed as an alternative treatment method. Bacteria can develop a resistance for phages, often having a negative effect on bacterial fitness. Acinetobacter baumannii is a priority pathogen of World Health Organization, developing antibiotic resistance easily and causing serious opportunistic infections in hospitals. In this bachelor's thesis, the effects of phage resistance on growth characteristics of Acinetobacter baumannii were studied. Bacterial strains from patients were exposed to phages capable of infecting them. The strains with developed phage resistance and strains not exposed to phages were then compared spectrophotometrically by Bioscreen C measurements. The development of phage resistance was detected to mainly weaken bacterial growth. Maximum bacterial population was reduced with every phage resistant strain and maximum growth rate was reduced with almost every phage resistant strain compared to its respective unexposed strain. The inhibition in growth is connected to, for example, decline in virulence and increased antibiotic sensitivity. As such, the results of this study bring promise towards phage therapy and antibiotic combination treatment development.
