Vaikka \nperim\u00e4ll\u00e4 ja elintavoilla voi olla haittavaikutuksia aivoterveyteen, voi\nhippokampuksen plastisuutta ja t\u00e4ten my\u00f6s kognitiivisia toimintoja mahdollisesti \nedist\u00e4\u00e4 lis\u00e4\u00e4m\u00e4ll\u00e4 neurogeneesi\u00e4 eli uusien hermosolujen synty\u00e4\nhippokampuksessa. T\u00e4ss\u00e4 LuK-tutkielmassa tutkittiin perityn juoksukapasiteetin ja \nliikunnan vaikutusta aikuisi\u00e4n hippokampaaliseen neurogeneesiin valikoidusti \njalostetun rottamallin avulla. Tutkittavat joko hyv\u00e4n tai huonon perityn \njuoksukapasiteetin omaavat rotat jaettiin juokseviin ja liikkumattomiin ryhmiin, \njotta voitiin selvitt\u00e4\u00e4 kuinka aerobinen liikunta lis\u00e4\u00e4 neurogeneesi\u00e4.\nLiikkumattoman ryhm\u00e4n avulla saatiin my\u00f6s tietoa siit\u00e4, kuinka pelkk\u00e4 peritty \njuoksukapasiteetti vaikuttaa neurogeneesiin. Lis\u00e4ksi huonon perityn \njuoksukapasiteetin rotat olivat perim\u00e4lt\u00e4\u00e4n alttiita ylipainolle ja metabolisille \nsairauksille. Rottien aivoleikkeiden uudet hermosolut v\u00e4rj\u00e4ttiin\nimmunohistokemiallisin menetelmin solujen Ki-67-proteiiniin sitoutuvalla vasta aineella. V\u00e4rj\u00e4tyt solut laskettiin sek\u00e4 kuvattiin valomikroskoopilla, ja solulaskujen \ntulokset analysoitiin tilastollisin menetelmin. Tutkimuksen tulosten mukaan \njokaisella rottaryhm\u00e4ll\u00e4 ilmeni neurogeneesi\u00e4, mutta ainoastaan huonon perityn \njuoksukapasiteetin omaavien rottien liikkuvan ryhm\u00e4n uusien jakautuneiden \nhermosolujen m\u00e4\u00e4r\u00e4 oli tilastollisesti merkitsev\u00e4 verrattuna liikkumattomaan \nryhm\u00e4\u00e4n. Tulokset osoittivat, ett\u00e4 aerobisella liikunnalla voitiin edist\u00e4\u00e4 \nneurogeneesi\u00e4 rottien huonosta perityst\u00e4 aerobisesta kunnosta huolimatta. Like aging, these risks have been shown to have a reducing effect on brain \nhealth and cognitive functions. In addition to lifestyle choices, such as physical \nexercise, genetics can have an impact on the susceptibility to these lifestyle related \ndiseases and also on brain health. Hippocampus, a structure located in the temporal \nlobe of the brain, is responsible for learning, memory, and emotional regulation. \nEven though genetics and lifestyles can have a debilitating effect on brain health, it \ncould still be possible to promote hippocampal plasticity, and therefore cognitive \nfunctions, by increasing neurogenesis, the formation of new neurons in the \nhippocampus. In this Bachelor's thesis the effect of inherited running capacity and \nexercise on adult hippocampal neurogenesis was studied by using a selectively bred \nrat model. These rats with either high or low inherited running capacity were \ndivided into running and sedentary groups to determine how aerobic exercise \ninduces neurogenesis. The sedentary group provided information on how the \ninherited running capacity alone affects neurogenesis. The rats with low inherited \nrunning capacity were also more prone to obesity and metabolic diseases. New \ndivided neurons in the rat hippocampus were stained immunohistologically with \nKi-67 antibody and counted under a light microscope. The results of cell counts \nwere analyzed by statistical methods. Each rat group showed signs of neurogenesis \nbut the only statistically significant result was shown between running and \nsedentary rat groups with low inherited running capacity, where the running group \nhad more new neurons in the hippocampus.
