Acinetobacter \nbaumannii on noussut esiin merkitt\u00e4vimp\u00e4n\u00e4 monen antibioottiresistanssin \nomaavana patogeenina, varsinkin sairaalaymp\u00e4rist\u00f6iss\u00e4. T\u00e4m\u00e4 johtuu p\u00e4\u00e4osin A. \nbaumanniin tehokkaista geneettisist\u00e4 ominaisuuksista, kuten sis\u00e4isen puolustuksen \nuudelleenj\u00e4rjestelyst\u00e4, jonka avulla bakteeri on kyennyt muun muassa ker\u00e4\u00e4m\u00e4\u00e4n \nresistenssisyyksi\u00e4 monia antibiootteja vastaan. Se my\u00f6s kykenee selvi\u00e4m\u00e4\u00e4n \nerilaisissa olosuhteissa, t\u00e4ten lis\u00e4ten sen levi\u00e4v\u00e4isyytt\u00e4. Tutkimuksen tavoitteena oli erist\u00e4\u00e4 faageja \nj\u00e4tevedest\u00e4 nelj\u00e4\u00e4 eri antibioottiresistentti\u00e4 A. baumannii kantaa vastaan, tutkia \nfaagiresistenssin kehityst\u00e4 ja m\u00e4\u00e4ritt\u00e4\u00e4 resistenssiyden vaikutukset bakteerin \nkasvuominaisuuksiin. Faagieristys j\u00e4tevedest\u00e4 ei onnistunut useista yrityksist\u00e4 \nhuolimatta. T\u00e4ten, seitsem\u00e4n aikaisemmin eristetty\u00e4 faagia karakterisoitiin \ntutkimuksessa plakkianalyyseill\u00e4, kasvukokeilla, DNA sekvensoinnilla ja elektronimikroskopoinnilla. Acinetobacter \nbaumannii has arisen as most notable multi-drug resistant species in clinical care \nassociated infections. This is due to A. baumannii\u2019s efficient genetic machinery and \nupregulation of innate resistance mechanisms, which has allowed the accumulated \nresistance towards multiple classes of antibiotics. Furthermore, it has remarkable\ncapability to survive harsh environments, which makes it possible for the bacteria \nto endure a long time and transmit to a possible host. New strains of A. baumannii\nhave emerged, which have acquired resistance towards almost all known classes of \nantibiotics. With increasing number of reports about multi-drug resistant A. \nbaumannii infections and outbreaks, new therapeutic approaches are needed to be \npursued. Using viruses that target bacteria (phages) to combat against bacterial \ndrug resistance is a potential therapy option. The aims of the study were to isolate \nphages from sewage water against 4 different antibiotic resistant A. baumannii\nstrains, assess the phage resistance mechanisms of development and determine \ntheir viability of phage therapy. Phage isolations were attempted from wastewater \nwithout success. Thus, 7 previously isolated phages from the same wastewater \nsource, were characterized via plaque assays, growth tests, genome sequencing and \nelectron microscopy.
