T\u00e4m\u00e4n pro gradu -tutkielman tarkoituksena oli selvitt\u00e4\u00e4 ulosteensiirron vaikutuksia LCR- (low- capacity runner, matalan aerobisen kapasiteetin juoksija) ja HCR (high-capacity runner, korkean aerobisen kapasiteetin juoksija) -rottien suolistomikrobiston koostumukseen, vapaaehtoiseen juoksuharjoitteluun, maksimisuorituskykyyn ja aineenvaihduntaa kuvastaviin veriarvoihin.\nT\u00e4ss\u00e4 tutkimuksessa k\u00e4ytetyt rotat olivat aerobiselta kapasiteetiltaan eroavia 44. sukupolven LCR- ja HCR -rottia, joita edelt\u00e4v\u00e4 sukupolvi kuljetettiin Suomeen Toledon yliopistosta Yhdysvalloista lokakuussa 2019. Rotat (n=32) jaettiin yhteens\u00e4 kahdeksaan ryhm\u00e4\u00e4n ja kaikille rotille tehtiin suoliston puhdistus mikrobeista suolihuuhtelun avulla. Nelj\u00e4lle ryhm\u00e4lle tehtiin lis\u00e4ksi ulosteensiirto. Suolistomikrobiston koostumusta tarkasteltiin tutkimuksen aikana kolmesti ja nelj\u00e4n ryhm\u00e4n osalta mitattiin h\u00e4kiss\u00e4 sijaitsevan juoksupy\u00f6r\u00e4n k\u00e4ytt\u00f6\u00e4. Tutkimuksen p\u00e4\u00e4tteeksi jokaisen ryhm\u00e4n maksimisuorituskyky\u00e4 mitattiin juoksumatolla. Verin\u00e4ytteet ker\u00e4ttiin lopetuksen yhteydess\u00e4.\nKaikkien rottaryhmien suolistomikrobiston koostumus vaikutti p\u00e4\u00e4jaksotasolla muotoutuvan kohti runsaampaa bakteroidi- p\u00e4\u00e4jakson ja niukempaa firmikuutti -p\u00e4\u00e4jakson osuutta, mutta muutokset eiv\u00e4t olleet merkitsevi\u00e4. Ainoastaan ulosteensiirron vastaanottaneen HCR-ryhm\u00e4n firmikuutti -p\u00e4\u00e4jakson osuuden havaittiin laskeneen (p<0.05) suolihuuhtelun saaneeseen LCR-ryhm\u00e4\u00e4n n\u00e4hden ulosteensiirron j\u00e4lkeen. Ulosteensiirron vastaanottaneen juoksupy\u00f6r\u00e4llisen HCR-ryhm\u00e4n LDL-kolesterolin m\u00e4\u00e4r\u00e4n todettiin lis\u00e4ksi olleen alhaisempi suolihuuhtelun saaneeseen LCR- (p<0.001) ja juoksupy\u00f6r\u00e4lliseen LCR -ryhm\u00e4\u00e4n (p<0.05) n\u00e4hden. Kokonaisjuoksumatkaa tarkastellessa ulosteensiirron vastaanottanut juoksupy\u00f6r\u00e4llinen HCR -ryhm\u00e4 juoksi tutkimuksen viimeisell\u00e4 viikolla enemm\u00e4n kuin ulosteensiirron vastaanottanut juoksupy\u00f6r\u00e4llinen LCR-ryhm\u00e4 (p<0.05). Kaikkien HCR-ryhmien maksimisuorituskyky, lukuun ottamatta ainoastaan suolihuuhtelun vastaanottanutta ryhm\u00e4\u00e4, todettiin suolihuuhtelun saanutta LCR-ryhm\u00e4\u00e4 paremmaksi (p<0.05).\nT\u00e4m\u00e4n pro gradu -tutkielman tulosten pohjalta voidaan todeta suolihuuhtelun ja LCR-rotilta saadun ulosteensiirteen v\u00e4hent\u00e4v\u00e4n mahdollisesti HCR-rottien suolistomikrobiston sis\u00e4lt\u00e4m\u00e4n firmikuutti -p\u00e4\u00e4jakson osuutta. Suolihuuhtelun ja ulosteensiirron avulla voidaan lis\u00e4ksi mahdollisesti v\u00e4hent\u00e4\u00e4 LCR -rottien seerumin LDL-kolesterolin m\u00e4\u00e4r\u00e4\u00e4 HCR-ryhmiin n\u00e4hden. Ulosteensiirron ei puolestaan voida t\u00e4m\u00e4n tutkielman tulosten perusteella katsoa vaikuttavan HCR- tai LCR -rottien maksimisuorituskykyyn tai vapaaehtoiseen juoksuharjoitteluun. The purpose of this master\u2019s thesis was to study the impact of a fecal microbiota transplantation on LCR- (low-capacity runner) and HCR (high-capacity runner) rats\u2019 gastrointestinal microbiota composition, running distance, physical performance, and metabolism-related serum variables.\nThis study examined 44th generation LCR- and HCR rats that have different aerobic capacity. The previous generation was transported to Finland from the University of Toledo (the United States of America) in October 2019. Rats (n=32) were divided into eight groups and their bowels were cleansed from microbes. Four groups were treated with a fecal microbiota transplant (FMT). The composition of the gastrointestinal microbiota of the rats was examined three times during the five-week study and voluntary running distance was measured from four groups which had running wheels in their cages. At the end of the study each group was tested for the maximum running capacity -test on the treadmill. Blood samples were collected at the time of sacrificing of the animals.\nIn all groups the composition of the gastrointestinal microbiota seemed to take shape towards the more abundant level of Bacteroidetes phylum and less abundant level of Firmicutes phylum although the shifts were not significant. Only the FMT-treated HCR-group was found to have decreased level of Firmicutes -phylum (p<0.05) after the bowel cleansing and FMT when compared to the LCR-group which underwent bowel cleansing only. The FMT-treated HCR-group with a running wheel also had a significantly lower LDL-cholesterol levels compared to the bowel cleansing -treated LCR-group (p<0.001) and LCR-group with a running wheel cage (p<0.05). When looking at the results of total running distance, the FMT -treated HCR-group ran significantly more (p<0.05) compared to the FMT-treated LCR-group with a running wheel cage. With the exception of bowel cleansing only -group, all HCR-groups had also significantly (p<0.05) better physical performance compared to the LCR-group that underwent bowel cleansing only.\nBased on the results of this study it can be stated that bowel cleansing and fecal microbiota transplantation from LCR-rats could possibly decrease the abundance of the firmicutes -phylum in the gastrointestinal microbiota of HCR-rats. Bowel cleansing and fecal microbiota transplantation could also possibly decrease the amount of LDL- cholesterol serum levels in LCR rats compared to HCR rats. However, based on the results of this study it cannot be stated that fecal microbiota transplantation would affect the physical performance or running distance of LCR- or HCR -rats. To support these conclusions a larger sample size and control groups are needed.
