Tutkimuksessa selvitettiin, onko beta-sitosterolilla vaikutusta geenien sytokromi P4501A (cyp1a), estrogeenireseptori alfa (erα) ja vitellogeniini (vtg) ilmentymiseen poikasissa. Lisäksi vaikutuksia poikasten pituuksiin ja ruskuaispussin pinta-aloihin tutkittiin. Erα ja vtg ilmentävät aineen estrogeenista vaikutusta ja cyp1a dioksiinityyppistä toksisuutta. Tutkimuksessa suoritettiin kaksi altistuskoetta pituuksiltaan 24 h ja 14 vrk. Beta-sitosterolin pitoisuudet olivat 1, 10, 100 ja 1000 μg/l, sekä positiivisena kontrollina toimi reteeni (100 μg/l). Reteenin liuotinkontrollina oli DMSO (10 μg/l) ja beta-sitosterolin liuotinkontrollina etanoli (20 ja 200 μg/l). Kummassakin kokeessa oli kolme toistoa kustakin pitoisuudesta, sekä 12 poikasta jokaisessa 24 vuoassa. 24 h näytteet tutkittiin reaaliaikaisen polymeraasiketjureaktion (qPCR) avulla, joka kertoo geenin lähetti-RNA:n määrän ja siten geenin ilmenemisen. Beta-sitosterolilla ei havaittu vaikutusta tutkittuihin geeneihin, eikä pituuksiin ja ruskuaispussien pinta-aloihin. Johtopäätöksenä voidaan todeta, ettei beta-sitosteroli aiheuta dioksiinin kaltaista toksisuutta järvitaimenen poikasissa, eikä sillä näyttäisi myöskään olevan estrogeenivaikutusta vastakuoriutuneisiin järvitaimeniin. Beta-sitosterol is an estrogenic compound which naturally occurs in trees. Pulp and paper mill effluents are major sources of beta-sitosterol from wood to water systems. Effects of beta-sitosterol have mainly been studied by using adult fish, so in this study objects of interests are possible effects on newly hatched fish larvae. The test species in the study was brown trout (Salmo trutta lacustris) that is a native species in Finland and may suffer from beta-sitosterol. In this survey we tested could beta-sitosterol affect the number of transcripts of cytochrome P4501A (cyp1a), estrogen receptor alpha (erα) and vitellogenin (vtg) in larvae. It was also investigated whether beta-sitosterol affect the lengths and yolk sac areas of larvae. Erα and vtg express an estrogenic effect of compound and cyp1a dioxin-like toxicity. In this study we had two experiments: 24 hours long and 14 days long. The concentrations of beta-sitosterol were 1, 10, 100 and 1000 μg/L and the positive control was retene (100 μg/L). Solvent control of retene was DMSO (10 μg/L) and solvent control of beta-sitosterol was ethanol in concentrations 20 and 200 μg/L. There were three replicates of each solution and in one test experiment there were 24 glass bowls and in each of these were 12 larvae. 24 hours samples were analyzed by using quantitative polymerase chain reaction (qPCR). Amounts of messenger RNA (mRNA) of different genes tell us the activity of the expression of these genes in the cells. Beta-sitosterol did not have any effects on the genes studied nor on the lengths and yolk sac areas. In conclusion it can be said that beta-sitosterol has no dioxin-like or estrogenic effects on newly hatched brown trout larvae.
