Kirjolohen (Oncorhynchus mykiss) tiedet\u00e4\u00e4n metaboloivan PAH:it sytokromi P450-entsyymij\u00e4rjestelm\u00e4\u00e4n (CYP450) kuuluvan CYP1A\n-entsyymin katalysoimilla hapetus-pelkistysreaktioilla. CYP1A-entsyymi\u00e4 s\u00e4\u00e4telee\naryylihiilivetyreseptori (AhR). Joidenkin PAH:ien tiedet\u00e4\u00e4n olevan AhR aktivaattoreitta ja lis\u00e4\u00e4v\u00e4n CYP1A-entsyymien toimintaa. T\u00e4ss\u00e4 ty\u00f6ss\u00e4 k\u00e4ytetyist\u00e4 yhdisteist\u00e4\nreteeni (Ret, 1-metyyli-7-isopropyylifenantreeni) on vahva AhR aktivaattori ja pyreeni (Pyr) ja fluoranteeni (Flu) ovat heikkoja aktivaattoreita. Flu on my\u00f6s CYP1Aentsyymin inhibiittori. T\u00e4ss\u00e4 Pro gradu -ty\u00f6ss\u00e4 tutkitaan, voiko yhteisaltistus\nCYP1A-entsyymituotantoa lis\u00e4\u00e4vien AhR-agonistien Ret tai Pyr kanssa v\u00e4hent\u00e4\u00e4\nmy\u00f6s CYP1A-inhibiittori Flu konsentraatiota. Ty\u00f6n aineisto on per\u00e4isin aikaisemmasta tutkimuksesta, jossa kirjolohen poikasia altistettiin 1, 3 ja 7 p\u00e4iv\u00e4n ajan yhdisteille Flu (5, 50 ja 500 \u00b5g/l), Ret ja Pyr (3,2; 10 ja 32 \u00b5g/l) tai seoksille Pyr+Flu ja\nRet+Flu (kaikki konsentraatio yhdistelm\u00e4t). T\u00e4ss\u00e4 tutkimuksessa poikaset homogenoitiin ja yhdisteiden pitoisuudet mitattiin korkean erotuskyvyn nestekromatografilla (HPLC) ja fluoresenssidetektorilla. Sek\u00e4 Pyr- ett\u00e4 Ret-pitoisuudet kasvoivat Flualtistuksessa. Rainbow trout (Oncorhynchus mykiss) is known to\nmetabolize PAHs through CYP1A enzymes that catalyze redox reactions. The\nCYP1A enzymes are part of the cytochrome P450 superfamily and are induced\ntrough aryl hydrocarbon receptors (AhR). Some PAHs are AhR agonists and induce\nCYP1A expression. Of the compounds used in this thesis retene (Ret, 1-methyl-7-\nisopropyl phenanthrene) is a strong AhR agonist and pyrene (Pyr) and fluoranthene\n(Flu) are weak. Flu is also CYP1A-enzyme inhibitor. The purpose of this thesis is to\ndetermine if co-exposure with CYP1A inducers Pyr or Ret will alter Flu tissue concentrations. Rainbow trout fry, from a previous study, were used as material in this\nthesis. They were exposed for 1, 3 or 7 days to Flu (5, 50 and 500 \u00b5g/l), Ret, Pyr (both\n3,2; 10 and 32 \u00b5g/l), Pyr+Flu or Ret+Flu (all the concentration combinations). The\nfry were homogenized and the tissue concentrations were measured with high-performance liquid chromatography (HPLC) and fluorescence detector. Both Pyr and\nRet body burdens increased with Flu exposure. Pyr exposure did not affect Flu body\nburden while coexposure with Ret resulted in some decrease over time.
