Siksi tarve uudentyyppisille stafylokokki-infektioita hoitaville l\u00e4\u00e4keaineille onkin kova. Bakteerien solusein\u00e4m\u00e4\u00e4 pilkkovat entsyymit \u2013 peptidoglykaanihydrolaasit \u2013 ovat er\u00e4s lupaavimmista uudentyyppisist\u00e4 antistafylokokkaalisista l\u00e4\u00e4keaineista. Kuten muillakin gram-positiivisilla bakteereilla, S. aureuksella on paksu bakteerisolua suojaava peptidoglykaani-sein\u00e4m\u00e4. Peptidoglykaanihydrolaasit ovat entsyymej\u00e4, jotka pilkkovat tiettyj\u00e4 kemiallisia sidoksia t\u00e4ss\u00e4 solusein\u00e4m\u00e4ss\u00e4, aiheuttaen siten bakteerisolun tuhoutumisen. Peptidoglykaanihydrolaasien keskeisimm\u00e4t hy\u00f6dyt ovat niiden (1) spesifisyys, sill\u00e4 entsyymit toimivat kohdennetusti pilkkoen juuri tiettyj\u00e4 kemiallisia rakenteita bakteerin peptidoglykaani-solusein\u00e4m\u00e4ss\u00e4, (2) matala myrkyllisyys, (3) entsyymien suhteellisen helppo ja tehokas tuotto, (4) niiden kyky tuhota sek\u00e4 aktiivisia ett\u00e4 inaktiivisia bakteerisoluja ja toimia my\u00f6s biofilmeiss\u00e4, sek\u00e4 lopuksi (5) entsyymien k\u00e4ytt\u00f6 antibioottien kanssa yhdistelm\u00e4hoitona. T\u00e4ss\u00e4 tutkielmassa tarkastellaan erilaisten synteettisten peptidoglykaanifragmenttien pilkkoutumista kahden peptidoglykaani hydrolaasin toimesta. Tutkittavat entsyymit kuuluvat lysostafiini-entsyymiperheeseen. Entsyymien tehokkuutta erilaisten peptidoglykaanifragmenttien katalyysiss\u00e4 tutkittiin k\u00e4ytt\u00e4en liuostilan 1D 1H ydinmagneettista resonanssispektroskopiaa (NMR), joka mahdollistaa hydrolyysireaktioiden reaaliaikaisen seurannan atomiresoluution tasolla. Reaktiotuotteiden rakenteet m\u00e4\u00e4ritettiin 2D NMR -spektroskopialla. Kummallekin tutkittavista entsyymeist\u00e4 l\u00f6ydettiin uusia substraatteja eli bakteerien peptidoglykaanirakenteita, joita ne pilkkovat kohdennetusti.
