Tavoitteenani oli tunnistaa ABC-geenejä koloradonkuoriaisen perimästä, sekä selvittää, ovatko ne yhteydessä sen torjunta-ainevastustuskykyyn. Tutkin 1) vaikuttavatko atsinfossimetyyli (AZ; organofosfaatti) ja karbaryyli (CAR; karbamaatti) ABCgeenin ilmentymiseen, sekä 2) eroaako niiden ilmentyminen torjunta-aineille vastustuskykyisen (Vermont, U.S.A) ja alttiimman (Belchow, Puola) populaation välillä. 18 yksilöä (yht. 36) kummastakin populaatiosta altistettiin joko torjuntaaineelle tai asetonille (kontrolli). Eristin RNA:n, valmistin komplementaarista DNA:ta ja analysoin kuuden ABC-geenin (ABCBmember6, ABCBmember7, ABCC13, ABCG1041A, ABCH278B, ABCH278C) ilmentymistasoja qPCRmenetelmällä. Sekä AZ että CAR voimistivat kaikkien ABC-geenien ilmentymistä. AZ-käsittelyn jälkeen ABCBmember6-geeni ilmentyi vastustuskykyisessä populaatiossa enemmän, kun taas ABCC13 ilmentyminen oli suurempaa alttiissa populaatiossa. Ilmentymisessä ei havaittu eroja populaatioiden välillä CAR- tai kontrollikäsittelyn jälkeen. Kaiken kaikkiaan ABC-geenit vaikuttavat olevan tärkeitä kuoriaisen suojaamisessa torjunta-aineilta. Etenkin ABCBmember6 on vahva ehdokas organofosfaattivastustuskyvyn osatekijäksi koloradonkuoriaisella. Insecticide resistance is a global issue caused by ever-increasing use of insecticides. One gene-family linked to the resistance is the family of ATP-binding cassette (ABC) transporters which e.g. excrete toxins from the cytoplasm. The Colorado potato beetle (CPB), Leptinotarsa decemlineata, is a widespread pest species. CPB causes massive losses to for instance potato crops and is hard to control due to its high resistance to insecticides: the beetle is known to be resistant to 56 insecticides. My aim was to identify ABC genes in the genome of CPB and find out if they are linked to insecticide resistance. I wanted to know 1) if azinphos-methyl (AZ; organophosphate) and carbaryl (CAR; carbamate) influence the expression of ABC genes in CPB and 2) whether the expressions differ between resistant (Vermont, U.S.A.) and more susceptible (Belchow, Poland) populations. 18 individuals (total 36) from each population were treated with either an insecticide or acetone (control). I extracted their RNA, synthesized cDNA, and performed qPCR with primers of six different ABC genes (ABCBmember6, ABCBmember7, ABCC13, ABCG1041A, ABCH278B, ABCH278C). Exposure to both AZ and CAR elevated the expression rates of all ABC genes across the populations. After AZ treatment, ABCBmember6 was more upregulated in the resistant population, whereas ABCC13 was more upregulated in the sensitive one. The gene expressions did not differ between the populations in either CAR or control groups. My study supports the importance of ABC genes in protecting CPB against these insecticides. Especially ABCBmember6 is a probable candidate for being a part of organophosphate resistance in the beetle.
