SERS-ilmi\u00f6 perustuu pintaplasmoniresonanssi-ilmi\u00f6\u00f6n (SPR) ja sen keski\u00f6ss\u00e4 ovat substraatteina toimivat hopea- ja kultananopartikkelit. Ty\u00f6n kirjallisuusosassa k\u00e4ytiin kattavasti l\u00e4pi Raman-spektroskopiaan ja SERS-menetelm\u00e4\u00e4n liittyv\u00e4t keskeisimm\u00e4t kokonaisuudet, kuten matemaattinen tausta, h\u00e4iri\u00f6t ja laitteiston rakenne. SERS-menetelm\u00e4ss\u00e4 tarkasteltiin substraattien, hot spottien ja aggregaation merkityst\u00e4 ilmi\u00f6n kannalta. SERS-menetelm\u00e4n k\u00e4ytt\u00f6mahdollisuuksia v\u00e4riaineiden ja r\u00e4j\u00e4hdeaineiden tutkimuksissa tarkasteltiin kirjallisuudesta l\u00f6ytyneiden esimerkkien kautta. Ty\u00f6n kokeellisessa osassa, ennen varsinaisia SERS-tutkimuksia, sitraattipelkistetty\u00e4 hopeakolloidia valmistettiin substraatiksi. My\u00f6s kaupallista kultakolloidia kokeiltiin. Nanopartikkeleiden aggregoinnin vaikutusta SERS-ilmi\u00f6n ilmentymiseen tutkittiin kaliumnitraatin ja natriumkloridin avulla. Hopeakolloidin laatu varmistettiin mittaamalla siit\u00e4 UV-Vis -spektri ja sen nanopartikkelijakauman kokoa selvitettiin pyyhk\u00e4isyelektronimikroskoopilla kuvantamalla. Ty\u00f6h\u00f6n valittiin n\u00e4ytematriiseiksi v\u00e4riaineet ja r\u00e4j\u00e4hdeaineet. V\u00e4riaineiden SERS-tutkimus oli ty\u00f6n p\u00e4\u00e4painopiste ja n\u00e4ytteiksi valittiin punaiset spray- ja ty\u00f6kalumaalit sek\u00e4 kuulak\u00e4rkikynien ja tussien musteet. R\u00e4j\u00e4hdeaineet valittiin, koska niiden forensiset tutkimukset ovat haastavia perinteisill\u00e4 tutkimusmenetelmill\u00e4. Tutkittavaksi r\u00e4j\u00e4hdeaineeksi valittiin trinitrotolueeni (TNT) ja selvitettiin, voidaanko SERS-menetelm\u00e4\u00e4 hy\u00f6dynt\u00e4\u00e4 r\u00e4j\u00e4hdeainej\u00e4\u00e4mien tunnistamisessa laimeasta TNT-liuoksesta. Ty\u00f6ss\u00e4 pystyttiin todistamaan, ett\u00e4 SERS-menetelm\u00e4 toimii ja sit\u00e4 voidaan hy\u00f6dynt\u00e4\u00e4 edell\u00e4 mainittujen n\u00e4ytemateriaalien vertailututkimuksissa ja apuna materiaalin tunnistuksessa. SERS-menetelm\u00e4n toiminta on vahvasti riippuvainen tutkittavan molekyylin rakenteesta, jolloin kaikista tuntemattomista spray- ja ty\u00f6kalumaalin\u00e4ytteist\u00e4 ei pystytty mittaamaan SERS-spektri\u00e4. Kuulak\u00e4rkikynien ja musteiden spektrit onnistuttiin mittaamaan SERS:ll\u00e4. K\u00e4sittelem\u00e4tt\u00f6m\u00e4st\u00e4 TNT-liuoksesta ei saatu mitattua hyv\u00e4\u00e4 SERS-spektri\u00e4, mutta pH 13 -s\u00e4\u00e4detyst\u00e4 liuoksesta saatiin spektri mitattua. SERS-menetelm\u00e4 soveltui hyvin spray- ja ty\u00f6kalumaalien sek\u00e4 musteiden mittauksessa. Menetelm\u00e4n soveltuvuus osoittautui huonoksi TNT:n mittauksessa.
