PAH-yhdisteitä päätyy ympäristöön esimerkiksi laskeumien, hulevesien ja öljyonnettomuuksien takia. Monivierasainevastustuskyky (MXR) on proteiinipohjainen eliöiden kyky vastustaa haitallisten aineiden kertymistä soluihin. MXR-mekanismia esiintyy niin aito- kuin esitumaisilla eliöillä. MXR-mekanismin toiminta voi estyä erilaisten yhdisteiden vaikutuksesta, joita on niin luonnollisia kuin ihmisen valmistamiakin. Tässä työssä tutkittiin ovatko PAH-yhdisteet antraseeni, kryseeni ja pyreeni vesikirpun Daphnia magna ja harvasukamadon Lumbriculus variegatus MXR-mekanismin inhibitoreita. Kokeet suorittiin siten, että kumpikin koe-eliö altistettiin kullekin PAH-yhdisteelle erikseen. Altistuksen jälkeen mitattiin, kuinka paljon koe-eliön oli kertynyt fluoresoivaa merkkiainetta ja verrattiin tätä sekä normaalia että ihbitiotilannetta kuvastaviin kontroleihin. Tutkimuksessa havaittiin, että pyreeni poikkesi muista tukituista PAH-yhdisteistä. Harvasukamadolla ilmeni MXR-järjestelmän voimistumista. Vesikirppu eritti ulos jotakin pyreenin metaboliittia. Havainnot osoittavat, että mikään PAH-yhdisteistä ei ollut MXR-järjestelmän inhibiittori eli tutkitut PAH-yhdisteet eivät yksittäisinä aiheuttaisi haitallisten aineiden kertymistä tutkittujen eliöiden soluihin. Usually polycyclic aromatic hydrocarbons (PAHs) are considered harmful compounds. For example, some PAHs are known to lower the reproduction of some organisms. PAHs are formed by uncomplete burning processes, and crude oil also contains high concentrations of PAH compounds. PAH compounds end up in the environment via precipitations, storm waters and oil accidents. Multixenobiotics resistance (MXR) is a protein-based defend mechanism, which allows the organisms to prevent the accumulation of harmful substances in the cells. The MXR mechanism occurs in both eukaryote and prokaryote organisms. MXR mechanism can be inhibited by differend kind of compounds. Some of inhibitors are natural orgin and some ones are artificial. The aim of this work was studying if anthracene, chrysene and pyrene are inhibitors of MXR mechanism in water flea Daphnia magna and oligochaeta Lumriculus variegatus. In studyings both of test organisms were treated separatetly to each tested PAH compound in three different concen-trations and one group with only fluorescent compound and other group whit fluorescent and known inhibitor. After of test the amount of fluorecence was measured and compared between every treatment group. Pyrene differed from other PAHs in this study. Pyrene acted as an inducer of MXR in Lumriculus variegatus, and Daphnia magna secreted some unknown metabolite of pyrene. None of the tested PAHs were inhibitors to any of tested organisms, so they do not behave as chemosensitizer.
