AAP-bakteereita ei ole alettu tutkia aktiivisesti kuin vasta hyvin \u00e4skett\u00e4in, ja erityisesti niiden kyvyst\u00e4 k\u00e4ytt\u00e4\u00e4 fotosynteesi\u00e4, ei tiedet\u00e4 paljoa. T\u00e4m\u00e4n tutkimuksen keski\u00f6ss\u00e4 oli selvitt\u00e4\u00e4, mit\u00e4 olosuhteita vaaditaan joko BChl a:n, sek\u00e4 v\u00e4hemmiss\u00e4 m\u00e4\u00e4rin \u03b2-karoteenien, tuottamisen induktioon tai inhibitioon Sphingomonas glacialis-bakteerilajin kannassa S2U11. Tavoitteena oli my\u00f6s tutkia BChl- ja karoteenituotantojen v\u00e4list\u00e4, sek\u00e4 bakteerikasvun v\u00e4list\u00e4 mahdollista suhdetta. Fyysisten olosuhdevaatimusten selvitt\u00e4miseksi S2U11-kantaa kasvatettiin eri kasvuolosuhteissa (ravinnemieltymykset, sek\u00e4 plankton vai biofilmi-kasvutapamieltymys), mukaan lukien pH (5.9, 6.5 ja 7.0). Valovaatimusten selvitt\u00e4miseksi S2U11-kantaa kasvatettiin eri n\u00e4kyv\u00e4n valon ja l\u00e4hi-infrapunavalon (NIR) valoaallonpituuksia k\u00e4ytt\u00e4en tai pime\u00e4ss\u00e4, sek\u00e4 jatkuvassa valossa ett\u00e4 valo-pime\u00e4sykli\u00e4 k\u00e4ytt\u00e4en. Tulosten analyysi pohjautui mitattuihin absorbanssispektreihin sek\u00e4 absorbanssien maksimiarvojen keskiarvoihin perustuviin histogrammeihin. S2U11 kykenee k\u00e4ytt\u00e4m\u00e4\u00e4n BChl-tuotannossa eri ravinneyhdistelmi\u00e4, ja suosii kasvutapana biofilmi\u00e4. BChl:n tuotanto oli tehokkainta pH:ssa 5.9. Valo-olosuhteista BChl:n tuotanto indusoitui parhaiten pime\u00e4ss\u00e4 tai NIR-aallonpituuksien valossa, ja on tehokkainta valo-pime\u00e4sykli\u00e4 k\u00e4ytt\u00e4m\u00e4ll\u00e4. N\u00e4kyv\u00e4n valon aallonpituuksilla vaikutti olevan inhiboiva vaikutus BChl-tuotantoon, joko foto-oksidatiivisen vahingon tai valoreseptoreiden aiheuttaman s\u00e4\u00e4telyn johdosta. Tutkimuksen johtop\u00e4\u00e4telm\u00e4t antaisivat ymm\u00e4rt\u00e4\u00e4, ett\u00e4 S. glacialis-kanta S2U11 on, itse asiassa, AAP-bakteeri perustuen sen BChl:n tuotannon ilment\u00e4miin tunnuspiirteisiin. Lis\u00e4ksi olisi olemassa osoitusta sen puolesta, ett\u00e4 AAP-bakteerit k\u00e4ytt\u00e4isiv\u00e4t fotosynteesi\u00e4 avustavassa tarkoituksessa oligotrofisissa olosuhteissa (NIR) valon l\u00e4sn\u00e4 ollessa, ja jonka s\u00e4\u00e4telyst\u00e4 vastaavat p\u00e4\u00e4s\u00e4\u00e4nt\u00f6isesti kyseisen bakteerin monipuolinen valikoima valoreseptoreita.", "language": "fi", "element": "description", "qualifier": "abstract", "schema": "dc"}, {"key": "dc.description.abstract", "value": "Aerobic anoxygenic photosynthetic (AAP) bacteria are heterogeneous group of bacteria that possess bacteriochlorophyll (BChl) a-based photosynthesis machinery, and are capable of photosynthesis in the presence of oxygen. The AAPs have not been studied in earnest until relatively recently and much, especially concerning their photosynthesis ability, is still not known. The focus of this study was to explore the conditions that are required to induce or inhibit BChl a, and to a lesser degree, \u03b2-carotenes production in the bacterial species Sphingomonas glacialis strain S2U11. The possible relationship between BChl and carotenes production, and bacterial biomass production, was also assessed. As regards of physical conditions requirements, the S2U11 strain was tested for growth medium (nutrient preference, and plankton versus biofilm), and pH (5.9, 6.5, and 7.0). As regards of light requirements, the S2U11 were grown under a variety of wavelengths from the visible spectra and from the near-infrared (NIR) region, or in darkness, under continuous light, or under a light-dark cycle. The analysis of the results was based on acquired absorption spectra, and histograms of averaged sample absorption maxima values. The S2U11 are able to utilize a variety of nutrient sources for BChl production with varying success, and prefer to grow as biofilm. BChl production was most efficient in pH 5.9. The S2U11 bacteria showed best BChl production levels in darkness, or in light from the near-infrared (NIR) region of the spectra. The BChl production was its most efficient under a light-dark cycle. Light from the visible region of the spectra seemed to inhibit BChl production, either through photo-oxidative damage, or because of the regulation by photoreceptors. In conclusion, the results indicated that S. glacialis strain S2U11 is, indeed, an AAP bacterium as evident by the characteristics of their BChl production.
