Alaraajojen lihasvoiman ja voimantuottotehon on taas havaittu laskevan ik\u00e4\u00e4ntymisen my\u00f6t\u00e4 ja ennustavan toiminnanvajausten synty\u00e4. Telomeerien pituuden yhteytt\u00e4 alaraajojen lihasvoimaan ja voimantuottotehoon on tutkittu kuitenkin viel\u00e4 hyvin v\u00e4h\u00e4n. Tutkimuksen tarkoituksena oli selvitt\u00e4\u00e4, onko veren leukosyyttien telomeerien pituudella yhteytt\u00e4 alaraajojen lihasvoimaan ja voimantuottotehoon ik\u00e4\u00e4ntyneill\u00e4, 63\u201376-vuotiailla naisilla, ja selitt\u00e4v\u00e4tk\u00f6 ik\u00e4, painoindeksi, sairastavuus ja fyysinen aktiivisuus t\u00e4t\u00e4 yhteytt\u00e4. Lis\u00e4ksi tutkittiin, ennustaako telomeerien pituus alaraajojen lihasvoiman ja voimantuottotehon heikentymist\u00e4 ik\u00e4\u00e4ntyneill\u00e4 naisilla kolmen vuoden seurannan aikana.\n\nTutkimus on osa Finnish Twin Study on Ageing \u2013 tutkimusta (FITSA), jossa tutkitaan geneettisten ja ymp\u00e4rist\u00f6tekij\u00f6iden merkityst\u00e4 toiminnanvajauksien syntyyn. Tutkimukseen osallistui 386 63\u201376-vuotiasta naiskaksosta. Veren leukosyyttien telomeeripituus m\u00e4\u00e4riteltiin qPCR-menetelm\u00e4ll\u00e4 tutkimuksen alussa vuonna 2000. Polven isometrinen ojennusvoima sek\u00e4 voimantuottoteho mitattiin sek\u00e4 tutkimuksen alussa ett\u00e4 kolmen vuoden seurannan j\u00e4lkeen. Ojennusvoima mitattiin dynamometrill\u00e4 ja voimantuottoteho Nottingham Leg Extensor Power Rig \u2013laitteella. Analyysimenetelmin\u00e4 k\u00e4ytettiin Pearsonin korrelaatiokertoimen tulkintaa, lineaarista regressioanalyysia, toisen asteen regressioanalyysia, toistettujen mittausten varianssianalyysia ja katoanalyysissa T-testi\u00e4.\n\nVeren leukosyyttien telomeerien pituudella ei todettu olevan yhteytt\u00e4 alaraajojen lihasvoimaan (p=0.274) eik\u00e4 voimantuottotehoon (p=0.622). Tutkittavien alaraajojen lihasvoima ja voimantuottoteho eiv\u00e4t heikentyneet tilastollisesti merkitsev\u00e4sti (p>0.050) kolmen vuoden seurannan aikana. Telomeeripituudella ja ajalla ei havaittu my\u00f6sk\u00e4\u00e4n yhteisvaikutusta alaraajojen lihasvoimaan (p=0.100) eik\u00e4 voimantuottotehoon (p=0.588).\n\nTutkimustulokset eiv\u00e4t tue n\u00e4kemyst\u00e4, ett\u00e4 veren leukosyyttien telomeeripituus olisi ik\u00e4\u00e4ntymiseen liittyv\u00e4\u00e4 alaraajojen lihasvoiman tai voimantuottotehon laskua selitt\u00e4v\u00e4 biologinen mekanismi. It is not fully known, what are the biological mechanisms that cause decrease in strength and power during aging. Telomere length is considered to be a marker of biological aging and predictor of age-related illnesses and mortality. However, it is not known if and how telomere length is associated with lower extremity muscle strength and power. The purpose of the study was to investigate the association between leukocyte telomere length and muscle strength and power in lower extremities in older women and to study whether age, body mass index, morbidity and physical activity explains this potential association. In addition, it was investigated whether leukocyte telomere length predicts decrease in muscle strength and power over the three year follow-up period.\nThe study is part of the Finnish Twin Study on Ageing (FITSA), which investigates the role of genetic and environmental factors in the development of functional disadvantages. The study involved 386 twin sisters aged 63-76. Leukocyte telomere length was determined by quantitative polymerase chain reaction at the beginning of the study in the year 2000. Isometric knee extensor strength was measured by dynamometer and leg extensor power with the Nottingham Leg Extensor Power Rig at the beginning of the study and after the three year follow-up. The data was analyzed using the Pearson correlation coefficient, linear regression analysis, polynomial regression analysis, repeated measures analysis of variance and T-test for estimate the bias in the drop-out analysis.\nThere was no association between leukocyte telomere length and lower extremity muscle strength (p=0.274) and power (p=0.622). Strength and power did not change significantly during the three-year follow-up (p>0.050). Telomere length and time also showed no interaction with lower- extremities muscle strength (p=0.100) or power (p=0.588).\nThe results of this study do not support the hypothesis that leukocyte telomere length would be an age-related biological mechanism explaining the decrease in muscle strength and power in lower extremities. Further studies with telomere analysis in longitudinal study setups and larger study samples are needed to confirm or refute these findings. It is also possible that telomeres may have tissue specific associations with muscle strength and functional performance and therefore determining telomere length from muscle satellite cells in future studies might be useful.
