Perim\u00e4n on ajateltu selitt\u00e4v\u00e4n suuren osan havaitusta vaihtelusta, tosin tutkimusn\u00e4ytt\u00f6 t\u00e4st\u00e4 puuttuu. Tutkimuksissa fyysisen aktiivisuuden on havaittu vaikuttavan rasvanhapetuskykyyn ja rasvanhapetuskyvyn on havaittu olevan yhteydess\u00e4 aineenvaihdunnalliseen terveyteen. T\u00e4m\u00e4n tutkimuksen tavoitteena olikin kartoittaa rasvanhapetuskyvyn yhteyksi\u00e4 perim\u00e4\u00e4n, fyysiseen aktiivisuuteen ja insuliiniherkkyyteen.\r\n \r\nTutkimukseen rekrytoitiin 23 identtist\u00e4 mieskaksosparia suomalaisesta kaksoskohortista. Kaksosista 41 osallistui portaittaiseen kuormitustestiin, jossa MFO m\u00e4\u00e4ritettiin hengityskaasuanalyysin perusteella. Vapaa-ajan liikunta-aktiivisuus (LTPA) m\u00e4\u00e4ritettiin liikuntahaastattelujen ja Baecken kyselylomakkeen avulla. Kehonkoostumus arvioitiin DXAmittauksella. Insuliiniherkkyys (Matsuda indeksi) m\u00e4\u00e4ritettiin sokerirasituskokeen avulla ja tuloksista laskettiin AUC-arvot plasman insuliini- ja glukoosikonsentraatioille. Yhteens\u00e4 19 kaksosparin molemmat kaksoset osallistuivat kuormitustestiin ja n\u00e4ist\u00e4 9 parin todettiin eroavan merkitsev\u00e4sti toisistaan viimeisen kolmen vuoden vapaa-ajanliikunnan suhteen. \r\n\r\nMFO korreloi voimakkaasti kaikkien (r=0,72, p<0,001) ja LTPA:n suhteen eroavien (r=0,78, p=0,014) parien kaksosten v\u00e4lill\u00e4. MFO korreloi viimeisen 12 kuukauden LTPA:n (r=0,26, p=0,035) ja Baecken kyselyn tuloksen kanssa (r=0,40, p=0,023), mutta ei viimeisen kolmen vuoden LTPA:n kanssa. LTPA:n suhteen eroavien parien v\u00e4lill\u00e4 ei havaittu kokonaisuudessaan eroa rasvanhapetuskyvyss\u00e4, tosin kolmen kaksosparin kaksosten v\u00e4lill\u00e4 ero oli muita pareja suurempi. MFO korreloi insuliinin AUC-arvon (r=-0,42, p=0,015), mutta ei Matsuda indeksin kanssa. \r\n\r\nTutkimuksen tulos tukee ajatusta, ett\u00e4 perim\u00e4 vaikuttaa suuresti rasvanhapetuskykyyn. Fyysinen aktiivisuus oli yhteydess\u00e4 rasvanhapetuskykyyn, mutta syy-seuraussuhdetta ei pystytty osoittamaan. Mostly, variability has been thought to emerge from genetic level, however, evidence of this is lacking. In previous studies, physical activity has been shown to influence MFO and MFO seems to associate with metabolic health. Thus, the purpose of this study was to investigate connections between MFO and genetic component, physical activity, and insulin sensitivity. \r\n\r\nA total of 23 monozygotic male twin pairs were recruited from a Finnish twin cohort. Of these pairs, 41 subjects (age 34.4 \u00b1 1.4 years) participated in an incremental exercise test with gas exchange analysis to measure MFO. Leisure time physical activity (LTPA) level was determined with two separate interviews and Baecke questionnaire. Body composition was assessed with DEXA. Oral glucose tolerance test was used to measure insulin sensitivity (Matsuda index). Also, plasma glucose and insulin concentrations during the test were quantified with AUC values. Both co-twins from 19 pairs participated in the exercise test. Additionally, 9 of these pairs were determined long-term discordant for past 3-year LTPA. \r\n\r\nMFO showed high pairwise correlation in all twin pairs (r=0.72, p<0.001) and in the subset of twins long-term discordant for past 3-year LTPA (r=0.78, p=0.014). MFO correlated with past 12-month LTPA level (r=0.26, p=0.035) and Baecke questionnaire result (r=0.40, p=0.023) but not with past 3-year LTPA level. Pairwise analysis in twins long-term discordant for LTPA did not show an effect of LTPA on MFO. However, in three twin pairs intrapair difference in MFO was relatively higher. MFO correlated (r=-0.42, p=0.015) with AUC of insulin but not with Matsuda index. \r\n\r\nResults support the thought that MFO capacity is largely dictated by genetic factors. MFO associates with physical activity level, however, causal relationship could not be fully proven. Results suggest that responses in MFO to changes in physical activity may vary. Link between MFO and insulin sensitivity warrants further research, as association previously found by others could not be confirmed in this study.
