Erityisesti pohjoisen pallonpuoliskon talvi koettelee hyo\u0308nteisia\u0308, joista enemmisto\u0308 suojautuu kylmyydelta\u0308 fysiologisten muutosten avulla. Ta\u0308rkea\u0308 plastinen vaste on kylma\u0308akklimaatio, joka kasvattaa kylma\u0308nkesta\u0308vyytta\u0308 melko kylma\u0308lle la\u0308mpo\u0308tilalle altistumisen seurauksena. Huolimatta paljon tutkitusta fenotyyppisesta\u0308 vasteesta kylma\u0308akklimaation sukupuoliriippuvuus seka\u0308 geneettinen tausta tunnetaan huonosti, etenkin geenien vaihtoehtoisesti silmukoituvien varianttien tasolla. Ta\u0308ssa\u0308 tyo\u0308ssa\u0308 tutkimuslajiksi valittiin Drosophila montana sen huomattavan kylma\u0308nkeston, selva\u0308n kylma\u0308akklimaatiovasteen ja Drosophila-suvun hyvin tunnetun geneetiikan vuoksi. Fenotyyppinen kylma\u0308akklimaatiovaste todennettiin mittaamalla kriittinen terminen minimila\u0308mpo\u0308tila (CTmin), eika\u0308 sukupuolten va\u0308lilla\u0308 havaittu siina\u0308 eroa. Kontrolli- (19 \u00b0C) ja akklimaatio-olosuhteista (6 \u00b0C) kera\u0308tyista\u0308 naaras- ja koiraska\u0308rpa\u0308sista\u0308 (3 kustakin na\u0308ytteesta\u0308) sekvensoitiin RNA Illumina TruSeq \u2013protokollalla. Na\u0308in saadut 50\u201390 miljoonaa 150 ema\u0308sparin pituista sekvenssiparia muokattiin ja linjattiin D. montanan genomiin seka\u0308 ma\u0308a\u0308ritettiin aineistossa poikkeavasti ekspressoituvat geenit, transkriptit ja eksonit. Ekspressio oli poikkeava sukupuolten va\u0308lilla\u0308 45 %:lla geeneista\u0308, mutta kylma\u0308akklimaatio va\u0308hensi sukupuolten va\u0308lisia\u0308 eroja. Ekspressioltaan merkitseva\u0308sti poikkeavia geeneja\u0308 lo\u0308ydettiin koirailta 1448 ja naarailta 1374, ja na\u0308ista\u0308 521 esiintyi molemmilla sukupuolilla. Geeniekspressio transkriptitasolla oli samankaltaista kuin geenitasolla, mutta eksonitasolla naarailta lo\u0308ydettiin koiraita enemma\u0308n ekspressioltaan poikkeavia geeneja\u0308. Geenien kylma\u0308akklimaatiovaste oli samankaltainen sukupuolten va\u0308lilla\u0308 (korrelaatio 0,61), ja molemmilla sukupuolilla ekspressioltaan poikkeavissa geeneissa\u0308 samankaltaisuus oli viela\u0308 suurempaa (0,91). Na\u0308ma\u0308 molemmissa sukupuolissa kylma\u0308akklimaatioon merkitta\u0308va\u0308sti reagoivat geenit muodostavat joukon, joka selitta\u0308nee suurelta osin D. montana ka\u0308rpa\u0308sten kylma\u0308nkesta\u0308vyyden parantumisen. Geenien toiminnallisessa analyysissa\u0308 energian kohdentaminen, aineenvaihdunta ja stressireaktio olivat keskeisia\u0308, mutta myo\u0308s sukupuolisidonnaisia prosesseja lo\u0308ytyi, kuten seriini/treoniini-proteiini-kinaasit koirailla ja orgaanisten anioinien kuljetus naarailla. Osaa lo\u0308ydetyista\u0308 prosesseista (esim. WD40 toistojakso koirailla) ja kandidaattigeeneista\u0308 (esim. sodh-2 naarailla) ei ole aiemmin yhdistetty hyo\u0308nteisten kylma\u0308akklimaatioon. The winter in the northern hemisphere presents a challenge for insects, most of which need to cope with the cold with physiological modifications. An important plastic response to low temperature stress is the cold acclimation response, whereby an insect\u2019s cold tolerance is improved following an exposure to sub-lethal low temperatures. The phenotypic cold acclimation response is well characterized, but its genetic basis is less well known, especially at the level of the alternatively spliced transcripts of the same gene. Also, sex- specificity of cold acclimation at the transcriptome level has yet to be studied. To address these questions, Drosophila montana was chosen for study organism due to its cold- resistant nature, clear acclimation response and well-documented genetics of the genus. Phenotypic cold acclimation response was confirmed by determining critical thermal minimum (CTmin), but no difference between sexes was found. Three samples of both sexes kept at both control (19 \u00b0C) and cold-acclimation (6 \u00b0C) conditions were generated and then used for RNA-sequencing with Illumina TruSeq protocol yielding 50\u201390 million 150 bp long paired end reads per sample. Reads were trimmed and mapped to the D. montana genome, normalized and differentially expressed (DE) genes, transcripts and exons were determined. Sex-biased expression was found in 45 % of the genes, but cold acclimation mitigated sex-specific differences: genes that were significantly more upregulated in males in cold acclimation were mostly female-biased and vice versa. In the differential expression analysis, 1448 genes were DE in males and 1374 in females, of which 521 were shared. The number of DE genes at the transcript and exon level were similar in both sexes, but there was a surplus of DE genes at the exon level in females. The cold acclimation response was similar between sexes (correlation 0.61), and genes DE in cold acclimation in both sexes had even higher correlation (0.91). DE genes shared by both sexes likely constitute a key gene set mainly responsible for improving the cold tolerance. Functional enrichment analyses of these genes found that energy allocation, metabolism and stress response were most extensively represented. Additionally, sex-specific processes such as serine/threonine-protein kinase metabolism in males and sodium- independent organic anion transport in females were found. A few enriched clusters (e.g. WD40 repeat in males) and candidate genes (e.g. sodh-2 in females) were novel to insect cold acclimation.
