Vaikka enterovirusten sekä T1D:n kehittymisen välillä on selkeä yhteys, virusten T1D:n aiheuttavaa mekanismia ei vielä tunneta. Hypoteesimme oli, että enterovirusinfektio aiheuttaa hiiren haimakudoksen tuhoutumista ja voimme paikantaa viruksen kuoriproteiineja näistä kudoksista tehdyistä ohutleikkeistä vasta-aineiden sekä cryo-immuno-elektronimikroskopian avulla. Hiiren haimakudokset altistettiin kolmen päivän ajan Coxsackievirus B1:lle (CVB1) ja B3:lle (CVB3). Hiiren haimanäytteet tuotettiin ja toimitettiin JDRF nPOD-Virus yhteistyöryhmän toimesta (eläinkokeet hyväksytty eettisen toimikunnan toimesta: R13028). Tässä tutkimuksessa, haiman näytteet tutkittiin käyttäen läpäisyelektronimikroskooppia (TEM). TEMin avulla tutkittiin virusten aiheuttaman kudostuho ja virusten sijainti haimassa. Virukset leimattiin ohuilta jääleikkeiltä enterovirusten vasta-aineilla ja 10 nm ja 5 nm proteiini A. kullalla. Infektoituneet solut osoittivat apoptoosin merkkejä. Lisäksi havaittiin virusten aiheuttamia kalvovesikkeleiden muutoksia sekä tummia kalvorakenteita soluissa. Vasta-aineet tunnistivat infektoitujen solujen solulimasta syntyneet uudet virusproteiinit, kun ei-infektoiduissa näytteissä oli vain vähän taustasignaalia. Tulokset osoittavat, että CVB1 ja CVB3 aiheuttavat rakenteellisia muutoksia ja apoptoosia hiiren haimakudoksissa. Tulosten perusteella, virusta valmistui sekä eksokriini- että endokriinikudoksissa. Tämän tutkimuksen tuloksia voidaan hyödyntää tutkittaessa enterovirusten aiheuttamia haimavaurioita. Coxsackievirus group B (CVB) belongs to the family of Picornaviridae and to the genus of Enterovirus. Picornaviruses are small, approximately 30 nm in diameter, non-enveloped, spherical and icosahedral viruses. Recent studies have indicated that viruses belonging to the Coxsackievirus group B contribute to the emergence of type 1 diabetes (T1D). Although there is a correlation between the viral infections and T1D, the mechanism of the viral pathogenesis in the development of diabetes is not fully known yet. Our hypothesis was that enterovirus infection causes destruction in the mouse pancreatic tissue and we can detect the viral capsid proteins from the ultrathin sections with cryo-immuno-EM. Mice were exposed (3 days) to Coxsackievirus B1 (CVB1) and Coxsackievirus B3 (CVB3). Mice pancreas samples were provided and submitted by the JDRF nPOD-Virus Group (under ethical approvement R13028). In this study, pancreatic samples were examined using transmission electron microscopy (TEM). With TEM, the tissue damage level, caused by the viruses, and location of the viruses was examined. The viruses were labeled on ultrathin cryosections with anti-enterovirus antibodies and 10 nm and 5 nm protein A gold particles. The cells showed signs of apoptosis. In addition, modified membrane vesicles associated with viruses and virus induced dark membrane structures were observed. The antibodies detected newly formed virus particles and capsid proteins sensitively in the cytoplasm of infected cells with low background in non-infected cells. The results show that CVB1 and CVB3 cause structural changes and apoptosis in the pancreas tissue and viruses are produced in both exocrine and endocrine structures. The results of this study can be utilized while exploring the reasons to pancreatic damage caused by enterovirus.
