Esimerkiksi HDL-partikkelien p\u00e4\u00e4proteiinin, apolipoproteiini A1:n (ApoA1), on arveltu liittyv\u00e4n luurankolihassolujen toimintaan ja esiintyv\u00e4n my\u00f6s muissa kudoksissa, kuin maksassa tai seerumissa. T\u00e4ss\u00e4 tutkielmassa tarkasteltiin ApoA1-proteiinin sek\u00e4 oksidatiiviseen fosforylaatioon osallistuvien proteiinien ilmentymist\u00e4 luurankolihaskudoksessa fyysiselt\u00e4 aktiivisuudelta eroavilla kaksosilla. Lis\u00e4ksi tutkittiin, onko seerumin ApoA1-pitoisuudella yhteytt\u00e4 luurankolihasten soluhengitykseen osallistuviin proteiineihin.\r\n\r\nTy\u00f6ss\u00e4 k\u00e4ytettiin Jyv\u00e4skyl\u00e4n yliopiston TwinActive (2007) - tutkimukseen ker\u00e4tty\u00e4 kaksosaineistoa (n=20; aktiivisia=10 ja inaktiivisia=10) sek\u00e4 lihasn\u00e4ytteist\u00e4 analysoituja DNA geenisirutuloksia, proteiinien geenien ilmentymisen tarkasteluun. Lis\u00e4ksi vuosina 2014 \u2013 2016 j\u00e4ljell\u00e4 olevista lihasn\u00e4ytteist\u00e4 (n=13) tehtiin t\u00e4t\u00e4 tutkielmaa varten erillisi\u00e4 analyysej\u00e4 kvantitatiivista polymeraasiketjureaktiota sek\u00e4 Western blot - menetelm\u00e4\u00e4 hy\u00f6dynt\u00e4en. Tulosten tilastollinen analysointi toteutettiin IBM SPSS 22 - ohjelmistolla.\r\n\r\nKaksosparien vapaa-ajan liikunnallinen aktiivisuus erosi 30 vuoden ajan keskim\u00e4\u00e4rin 9 MET tuntia/p\u00e4iv\u00e4 (p\u22640,05). Mann Whitney U-testill\u00e4 arvioituna, seerumin ApoA1- ja ApoA1/ApoB-suhteessa ei havaittu eroja ryhmien v\u00e4lill\u00e4. Sen sijaan ApoA1-geeni ilmentyi kaksosten lihasn\u00e4ytteiss\u00e4 mRNA-tasolla, mutta ei eronnut tilastollisesti merkitsev\u00e4sti ryhmien v\u00e4lill\u00e4. Niin ik\u00e4\u00e4n ApoA1- ja HDL- partikkelien toimintaan liittyvien reseptorien (ABCA1, ABCG1, SR-B1, HNF\u03b1/\u03b3) ja entsyymien geenej\u00e4 (LCAT, CETP, PLPT, HL, PON1,2,3) ilmentyi kaksosten lihasn\u00e4ytteiss\u00e4 mRNA-tasolla.\r\n\r\nWestern blot - m\u00e4\u00e4rityksiss\u00e4 oksidatiiviseen fosforylaatioon liittyvien proteiinien tasoissa ei havaittu eroja ryhmien v\u00e4lill\u00e4 (n=13). Menetelm\u00e4 arvioi kuitenkin ainoastaan yhden proteiinin ilmentymist\u00e4 kussakin kompleksissa. Sen sijaan DNA geenisiruanalyysin perusteella 17 geenin ilmeneminen erosi tilastollisesti merkitsev\u00e4sti (p\u22640,05) ryhmien v\u00e4lill\u00e4 (n=20). Aktiivisilla kaksosilla geenien ilmentyminen n\u00e4ytti olevan tehokkaampaa etenkin kompleksin 1 proteiineissa kuin inaktiivisilla kaksosilla. Seerumin ApoA1- muuttujilla ei ollut yhteytt\u00e4 luurankolihaskudoksen proteiinim\u00e4\u00e4rityksiin (n=13).\r\n\r\nTutkielma osoitti, ett\u00e4 ApoA1-geeni\u00e4 sek\u00e4 ApoA1- ja HDL-partikkelien toimintaan liittyvien reseptorien ja entsyymien geenej\u00e4 ilmentyi luurankolihaskudoksissa ihmisill\u00e4. ApoA1 ilmentymisen taso oli kuitenkin v\u00e4h\u00e4ist\u00e4 verrattuna muiden t\u00e4ss\u00e4 ty\u00f6ss\u00e4 tutkittujen geenien ilmentymisen tasoihin. Niin ik\u00e4\u00e4n ApoA1 ilmentyminen proteiinitasolla sek\u00e4 toiminta luurankolihaksissa ihmisill\u00e4 j\u00e4iv\u00e4t ratkaisematta. Fyysisen aktiivisuuden erot n\u00e4yttiv\u00e4t vaikuttavan luurankolihasten oksidatiiviseen fosforylaatioon osallistuvien proteiinien ilmentymiseen mRNA-tasolla, samasta perim\u00e4st\u00e4 huolimatta. Fyysisell\u00e4 aktiivisuudella saattaa olla positiivisia vaikutuksia esimerkiksi ik\u00e4\u00e4ntymisen mukanaan tuomiin muutoksiin mitokondrioiden proteiinikompleksien rakenteissa. However recent studies have shown several other functions of HDL particles. For instance apolipoprotein A1 (ApoA1), the main protein of HDL particle has been associated with the components of skeletal muscle. Additionally ApoA1 have been shown to exist in other tissues than liver, or serum. The aim of this study was to investigate the gene expression of ApoA1 in human skeletal muscle. Furthermore the additional purpose of this study was to examine the expression of proteins involved in oxidative phosphorylation, and their association with serum ApoA1 in twin pairs discordant for leisure time physical activity.\r\n\r\nThe research data of TwinActive study (n=20; active=10 and inactive=10) which was conducted in the University of Jyv\u00e4skyl\u00e4 in 2007 were utilized in the present study. The participants volunteered to give muscle samples in 2007. DNA microarray results (n=20) which had been carried out in 2008 of these samples were utilized in this study. Additionally separate analyses of the remaining muscle samples (n=13) were carried out with quantitative polymerase chain reaction and Western blot methods. The statistical analysis was carried out utilizing IBM SPSS 22 - software.\r\n\r\nTwin pairs had been discordant for physical activity on average 9 MET hours/day (p\u22640,05) over 30 years. There were no difference between the groups in serum ApoA1 and ApoA1/ApoB ratio evaluated by Mann Whitney U - test. However ApoA1 gene was expressed in skeletal muscle samples of all participants at the mRNA level. The expression did not differ significantly between the groups. Additionally specific ApoA1 and HDL receptors (ABCA1, ABCG1, SR-B1, HNF\u03b1/\u03b3) and enzymes (LCAT, CETP, PLPT, HL, PON1,2,3) were expressed in skeletal muscles.\r\n\r\nWestern blot method did not reveal differences in the proteins of oxidative phosphorylation between the groups (n=13). However the method analysed only one protein of each complex. In contrast DNA microarray analysis showed that expression of 17 genes differed significantly between the groups (n=20). Especially genes coding the proteins of complex 1 appeared to be upregulated in the physically active twins (n=10). Serum ApoA1 variables were not associated with proteins of oxidative phosphorylation (n=13).\r\n\r\nThis study showed that the ApoA1 gene and genes involved in ApoA1 and HDL particles were expressed in human skeletal muscle. However the gene expression level of ApoA1 was low compared to the expression of other genes examined in this study. ApoA1 expression at the protein level as well as function in skeletal muscle in humans remained unresolved. Discordance in physical activity affected the expression of proteins involved in oxidative phosphorylation despite the identical or mainly identical genetic factors. Physical activity may have positive effects to the structures of protein complexes and it may delay age-dependent alterations in oxidative phosphorylation.
