N\u00e4m\u00e4 valoaktiiviset proteiinit muuntavat \r\nkonformaatiotaan palautuvasti aktiivisen (Pfr) ja inaktiivisen (Pr) tilan v\u00e4lill\u00e4. Fytokromien \r\naminohappojen v\u00e4liset heikot vuorovaikutukset - etenkin vetysidokset - vaikuttavat lopulta \r\nlaskostumiseen ja proteiinin l\u00e4mp\u00f6stabiiliuteen. Fytokromit esiintyv\u00e4t homodimeerisin\u00e4 \r\nrakenteina, joiden monomeerit koostuvat useista domeeneista. N\u00e4m\u00e4 jakautuvat N-\r\nterminaaliseen valosensitiiviseen moduuliin (PSM) ja C-terminaaliseen OutPut moduliin\r\n(OPM). PSM koostuu kolmesta domeenista (PAS-GAF ja PHY). T\u00e4ss\u00e4 ty\u00f6ss\u00e4 tutkittiin\r\nPSM:n eri konstruktioita UV-Vis spektroskopialla - kustakin sek\u00e4 dimeeri\u00e4 ett\u00e4 \r\nmonomeeri\u00e4 sek\u00e4 Pr, ett\u00e4 Pfr tiloissa. Spektrit mitattiin l\u00e4mmitt\u00e4en n\u00e4ytteit\u00e4 viiden asteen \r\nintervalleina 20 C:sta 85 C:een. Absorbanssin muutoksista l\u00e4mp\u00f6tilan funktiona p\u00e4\u00e4tteltiin\r\nkunkin n\u00e4ytteen l\u00e4mm\u00f6nkest\u00e4vyytt\u00e4. Havaittiin, ett\u00e4 verrattaessa dimeeri\u00e4 ja monomeeri\u00e4 \r\ndimeeri sieti noin viisi astetta enemm\u00e4n l\u00e4mmityst\u00e4. Kun verrattiin PAS-GAF-PHY:a ja \r\nPAS-GAF:a havaittiin, ett\u00e4 PAS-GAF-PHY siet\u00e4\u00e4 noin 10 astetta suurempaa l\u00e4mp\u00f6tilaa \r\ndenaturoitumatta merkitt\u00e4v\u00e4sti. Mittausten j\u00e4lkeen n\u00e4ytteet j\u00e4\u00e4hdytettiin \r\nhuoneenl\u00e4mp\u00f6tilaan ja niiden spektrit mitattiin uudestaan. These proteins change their conformation \r\nreversibly between active (Pfr) and inactive (Pr) state, in response to lightning conditions.\r\nThey were first discovered from plants in 1959, but despite intensive studies, the actual \r\nmechanism of their action still holds some mysteries. Weak interactions between the amino \r\nacid residues of phytochromes, alters in their thermal stability \u2013 mainly through hydrogen \r\nbonds. Phytochromes consists from a N-terminal photo sensing module (PSM) and from a \r\nC-terminal output module (OPM). The photo sensing module contains three domains (PAS-\r\nGAF and PHY). In this experiment the heat sensitivity of different photo sensing module \r\nconstructs were monitored (PAS-GAF-PHY dimer and monomer, and PAS-GAF dimer and \r\nmonomer) by UV-Vis spectroscopy. Measurements were done by heating various PSM \r\nconstructs in Pr and Pfr states. Spectra were measured at 5-degree intervals from 20 C to 85 \r\nC. The heat durability of samples were determined from the spectral changes. As a result,\r\ncomparison between a dimer and monomer showed higher heat tolerance in a dimeric \r\nconstruct. Comparison between PAS-GAF-PHY and PAS-GAF showed that PAS-GAF-\r\nPHY tolerated approximately 10 degrees higher temperature before denaturation. After \r\nspectral measurements samples were cooled and their spectra were measured. There was\r\nsome difference between cooled spectra of PAS-GAF-PHY and PAS-GAF samples.
