Tutkimus toteutettiin pelkoehdollistamisasetelmaa käyttäen. Ehdottomana ärsykkeenä toimi sähköisku ja ehdollisena ärsykkeenä 4kHz/70dB ääni. Tutkimuksessa oli 14 Sprague-Dawley urosrottaa. Elektrodien avulla kerätty tieto koottiin habituaation, klassisen pelkoehdollistamisen, vihje- ja kontekstitestien sekä ekstinktio asetelmien aikana. Testit videoitiin ja rottien pelkokäyttäytymistä mitattiin laskemalla aika, jonka rotat olivat jähmettyneinä ehdollisen ärsykkeen jälkeen. Theta-aktiivisuus oli korkeampaa ehdollistamisen aikana hippokampuksessa kuin striatumissa. Muuten testin aikana merkittävimmät theta-aktiivisuus tason muutokset olivat dorsaalisessa hippokampuksessa habituaation aikana sekä striatumissa ekstinktion aikana. Habituaation aikana hippokampuksessa ja ekstinktion aikana striatumissa theta-aktiivisuus oli ehdollisen ärsykkeen aikana matalampaa kuin ennen tai jälkeen ehdollisen ärsykkeen. Theta-aktiivisuus oli ekstinktion aikana matalampaa hyvien oppijoiden ryhmässä kuin huonojen oppijoiden ryhmässä. Theta-aktiivisuus on selvästi hippokampukselle tyypillistä aktiivisuutta ja erityisen merkittävää ennen ja jälkeen ehdollisen ärsykkeen, mutta ei ehdollisen ärsykkeen aikana. Striatumin theta-aktiivisuudella saattaa olla merkitystä ekstinktiolle. Kuten myös aiemmissa tutkimuksissa on havaittu, theta-aktiivisuus on erittäin tyypillistä hippokampukselle oppimisen kannalta. Striatumissa aktiivisuus on vähäisempää, mutta merkittävää mahdollisesti erityisesti pois-oppimisen aikana. The aim of this study was to investigate the changes in theta activity in dorsal hippocampus and striatum during fear conditioning. Theta oscillations are known to be typical for hippocampus and we wanted to get deeper information on what happens in hippocampus and striatum during fear conditioning and learning. Our unconditioned stimulus was an electric shock and the conditioned stimulus was a 4kHz/70dB tone. As our study subjects we had 14 Sprague-Dawley male rats. Electrodes were placed in the brains of the rats and information was gathered during habituation, classical conditioning, cue test, context test and extinction. Our experiments were video-taped and the fear response behaviour was analyzed by calculating the percentage of the time the rat spent frozen after the conditioned stimulus. Theta activity was higher in hippocampus than in striatum during fear conditioning. Otherwise the main changes in theta levels were in dorsal hippocampus during habituation and in striatum during extinction. During habituation in dorsal hippocampus and during extinction in striatum theta levels were lower during the conditioned stimulus than before of after the conditioned stimulus. Good and poor learners showed difference during extinction as the theta levels were lower among the good learner group. Theta activity is clearly typical for hippocampus and significant before and after the conditioned stimulus but not during the conditioned stimulus. Theta activity in striatum might play a more significant role in extinction. Like previous research has shown theta activity is greatly typical for hippocampus concerning learning. In striatum theta activity levels are lower during learning but probably more significant during extinction.
