Tieto siitä, miten aivojen oskillaatiot ovat sidoksissa käyttäytymiseen ja oppimiseen, on tärkeää tulevien sovellusten, kuten kuntoutuksen vuoksi. Aiheeseen liittyvät tutkimukset voisivat tarjota tärkeää informaatiota rappeuttavista aivosairauksista, kuten Alzheimerin taudista. Tutkimuksessamme käytimme Wistar -rottia, jotka jaettiin kolmeen ryhmään: välittömän stimulaation, viivästetyn stimulaation ja kontrolliryhmään. Pelkoehdollistamisen jälkeen välittömän stimulaation ja viivästetyn stimulaation ryhmille annettiin dentate-piikki riippuvaisia sähköisiä stimulaatioita angular bundle -alueelle. Sähköinen stimulaatio jäljitteli voimakkuudeltaan luonnollista dentate-piikkiä. Välittömän stimulaation ryhmä sai sähköärsykkeen heti, kun dentate-piikki ilmestyi. Viivästetyn stimulaation ryhmä sai sähköärsykkeen 300-600 millisekuntia dentate-piikin ilmestymisen jälkeen. Kontrolliryhmälle ei annettu sähköärsykettä. Konteksti- ja vihjetestit suoritettiin pelkoehdollistamisen jälkeisenä päivänä ja rottien jähmettyminen mitattiin manuaalisesti jälkeenpäin. Ryhmien välillä ei ilmennyt tilastollisesti merkitseviä eroja jähmettymisessä eli dentate- piikkien aikaisella stimuloinnilla tai viivästetysti esitetyllä stimuloinnilla ei ollut vaikutusta oppimiseen. Myöskään dentate-piikkien yleisyys ei ennustanut jähmettymistä välittömän ja viivästetyn stimulaation ryhmissä. Tulokset voivat johtua tutkimusasetelmasta tai siitä, että dentate-piikeillä ei ole vaikutusta rottien jähmettymiseen. Dentate-piikkejä koskevan vähäisen tiedon vuoksi dentate-piikkien yhteyksiä käyttäytymiseen tulisi tutkia laajemmin erilaisin menetelmin. n this research our aim was to study the effects of a dentate spike stimulation on learning. Previous studies have investigated how the dentate spikes affect the oscillatory dynamics in a hippocampal formation, but we could not find earlier studies that had focused on how the dentate spikes are related to visible behaviour. The knowledge about how brain oscillations are related to behaviour and learning is important for future adaptations, for instance rehabilitation, but also can enable better understanding of degenerative brain diseases such as Alzheimer’s disease. In this study we used male Wistar rats which were divided in three groups, in an immediate, a delay and a control group. After trace fear conditioning the immediate and the delay groups received the dentate spike contingent stimulation to angular bundle: the immediate group received stimulation simultaneously when the dentate spike was predicted to occur and the delay group received stimulation 300-600 ms after the dentate spike had occurred. The stimulation imitated the natural dentate spike. The control group did not receive any stimulation. The context test and cue test were performed the next day and the freezing of the rats were measured manually afterwards. The results did not show significant difference in freezing between groups, indicating that the dentate spike stimulation did not have an effect on learning. Also the frequency of the dentate spikes did not predict freezing in immediate and delay groups. This might be due to settings of our study or the possibility that the dentate spikes are not related to freezing behaviour in rats. Since little is known of the dentate spikes, the behavioral correlates could be studied with different settings. Also the effects of the dentate spikes to other brain oscillations should be studied more.
