Järjestelmän havaittiin soveltuvan heikosti erityisesti nopeiden molekyylien, kuten EGFP-proteiinin mittaamiseen. Suurin haitta oli ilmaisimen häiriösignaali, jonka seurauksena korrelaatioanalyysin tulokset poikkesivat merkittävästi kirjallisuusarvoista. Työssä tehtiin lisäksi yleisiä huomioita RICS-menetelmän yksityiskohdista. Esimerkiksi mikroskoopin skannausnopeuden muuttumisella todettiin olevan yllättävän suuri merkitys ottaen huomioon sen, ettei sitä tiettävästi ole käsitelty aiemmissa julkaisuissa. Raster image correlation spectroscopy (RICS) is a microscopy technique for quantitatively studying the dynamics of fluorescent species from 2D confocal images. In this thesis the theoretical and practical principles of RICS are discussed and demonstrated. On the basis of the theory, the applicability of a commercial confocal microscope system (Nikon A1R+, Nanoscience Center, University of Jyväskylä) for RICS diffusion measurements is determined. The system was found to be inefficient especially for measuring fast molecules, such as EGFP. The greatest drawback was high detector correlation, because of which results determined from fast fluctuations deviated significantly from literature values. Measurements were performed also to study the RICS method on a more general level. For example variable scan speed was found to be surprisingly important considering the fact that apparently it has not been discussed in previous publications.
