T\u00e4m\u00e4 tavanomaisesti spirokeettana esiintyv\u00e4 bakteeri kykenee vaihtamaan muotoaan ymp\u00e4rist\u00f6olosuhteiden muuttuessa ep\u00e4suotuisiksi. Borrelia-bakteerin eri esiintymismuotojen uskotaan olevan borrelioosin pitkittyneen taudinkuvan takana, mink\u00e4 vuoksi n\u00e4iden muotojen tutkiminen on t\u00e4rke\u00e4\u00e4. T\u00e4m\u00e4n pro gradu \u2013ty\u00f6n tavoitteena oli tutkia B. burgdorferin pleomorfisia muotoja ja l\u00f6yt\u00e4\u00e4 muotoja erottavia morfologisia tekij\u00f6it\u00e4 sek\u00e4 eroja proteiiniekspressiossa ja borrelioosipotilasseerumivasteessa spirokeettojen ja py\u00f6re\u00e4n esiintymismuodon v\u00e4lill\u00e4. Olettamuksena oli, ett\u00e4 konfokaalimikroskopialla muotojen v\u00e4lilt\u00e4 l\u00f6ydet\u00e4\u00e4n rakenteellisia eroja lipidi- ja hiilihydraattiarkkitehtuurissa. My\u00f6s proteiiniekspression ja borrelioosipotilaiden seerumivasteen oletettiin olevan yksil\u00f6llinen kullekin muodolle. Kollageeniin sitoutuva Acid Fuchsin \u2013v\u00e4ri v\u00e4rj\u00e4si ainoastaan biofilmej\u00e4, kun taas N-asetyyliglukosamiineihin sitoutuva WGA-v\u00e4ri (Wheat Germ Agglutinin) tarttui vain vedell\u00e4 aikaansaadun py\u00f6re\u00e4n esiintymismuodon (round body) pinnalle. WGA-v\u00e4rj\u00e4yksen tulos viittaa peptidoglykaanin paljastumiseen bakteerin ulkopinnalla py\u00f6re\u00e4ss\u00e4 esiintymismuodossa. L\u00e4p\u00e4isyelektronimikroskooppikuvista koottiin kuvasarja mallintamaan spirokeetan laskostumista py\u00f6re\u00e4\u00e4n esiintymismuotoon. Lis\u00e4ksi n\u00e4ist\u00e4 kuvista tarkasteltiin kaksoiskalvorakenteen ja flagellojen s\u00e4ilymist\u00e4 py\u00f6re\u00e4ss\u00e4 esiintymismuodossa ja havaittiin nelj\u00e4n p\u00e4iv\u00e4n inkuboinnin ihmisen seerumissa aiheuttavan muodon muutoksen py\u00f6re\u00e4\u00e4n esiintymismuotoon. Lis\u00e4ksi ihmisen seerumin havaittiin aiheuttavan runsasta vesikkelien erityst\u00e4 sek\u00e4 protoplasmisen sylinterin turpoamista. Flagellan s\u00e4ilyminen py\u00f6re\u00e4ss\u00e4 esiintymismuodossa todettiin lis\u00e4ksi immunoleimaamalla flagella p41 -vasta-aineella. Flagellan s\u00e4ilyminen ja vesikkeleiden eritys viittaavat siihen, ett\u00e4 py\u00f6re\u00e4 esiintymismuoto kykenee aiheuttamaan immuunivasteen. Lis\u00e4ksi WGA-v\u00e4rj\u00e4ys todisti ainakin toisen solusein\u00e4n p\u00e4\u00e4komponentin, N-asetyyliglukosamiinien, esiintymisen my\u00f6s py\u00f6re\u00e4ss\u00e4 esiintymismuodossa. As the disease develops it becomes more challenging to diagnose. Pleomorphic forms might be responsible for the prolonged disease profile, even though much controversy exists about them. Thereby the aim of this study was to examine the pleomorphism of B. burgdorferi. The hypothesis is that each pleomorphic form has specific morphological traits and unique antigenicity. Protein expression is also different in spirochetes and round bodies (RBs). The collagen dye Acid Fuchsin stained only biofilms whereas N-acetyl glucosamine dye wheat-germ agglutinin (WGA) stained RBs. WGA staining illustrated that one of the main components of bacterial cell wall, N-acetyl glucosamines, are still present in RBs and indicated that peptidoglycan would become exposed at the surface of RBs. Human serum induced RB formation suggesting that this form has physiological significance. Transmission electron microscope (TEM) images lead to a model demonstrating how the spirochetes could fold into RBs. The presence of flagella in the RBs was examined by immunolabeling with flagellar antibody p41. TEM images of human serum induced RBs revealed increased vesicle formation and swelling of protoplasmic cylinders. Undamaged phospholipid bilayers were observed in RB TEM images. In addition, protein profiles of spirochetes and H2O RBs were analyzed with two-dimensional polyacrylamide gel electrophoresis (2D PAGE). Analysis of the protein profiles did not reveal a decrease in the overall protein expression of RBs, which might have been expected if this form would have a damaged outer cell wall. Instead, an increase of the expression of several proteins of molecular weight 15-40 kDa was observed in the RBs. These proteins might be needed for adjusting to the unfavorable environmental conditions. Antigenicity of spirochetes and RBs were compared by western blots immunolabeled with Lyme patient sera. The blots demonstrated that RBs could induce an immune response. Moreover, the immune response is different from spirochetes. However, the antigen-antibody response seemed to vary greatly depending on the serum. Still, RBs could be clinically important since their antigenicity differs from the parent spirochete form.
