Erilaiset hyytym\u00e4n muodostamat veritulpat ovat merkitt\u00e4v\u00e4 kuolinsyy vuosittain.\nVerihiutaleet ovat keskeinen tekij\u00e4 t\u00e4ss\u00e4 tapahtumassa. Niiden aktivaatio tapahtuu kalvoreseptoreiden ja solun sis\u00e4isen signaloinnin monimutkaisesta vuorovaikutuksesta. T\u00e4m\u00e4n tutkimuksen kohteina ovat pernan tyrosiinikinaasi ja filamiini A, joiden v\u00e4linen vuorovaikutus on t\u00e4rke\u00e4\u00e4 verihiutaleiden toiminnalle. Verihiutaleet, joissa t\u00e4m\u00e4 vuorovaikutus on h\u00e4iriintynyt, ovat morfologisesti ep\u00e4normaaleja eiv\u00e4tk\u00e4 toimi normaalilla tavalla hyytym\u00e4n muodostumisessa.\nPernan tyrosiinikinaasi, Syk (engl. spleen tyrosine kinase) on ei-reseptorityyppinen solun signalointiin liittyv\u00e4 proteiinikinaasi. Se esiintyy solussa sek\u00e4 solulimassa ett\u00e4 tumassa. Syk on osallisena monissa solun fysiologisissa tapahtumissa, esimerkiksi morfogeneesiss\u00e4, solun kasvussa, migraatiossa ja solun tukirangan uudelleen j\u00e4rjest\u00e4ytymisess\u00e4.\nFilamiini on kookas solutukirangan proteiini. Filamiinin tiedet\u00e4\u00e4n vuorovaikuttavan useiden muiden proteiinien kanssa monissa eri rooleissa. Erityisen\u00e4 kysymyksen\u00e4 oli selvitt\u00e4\u00e4, vaikuttaako filamiinin nelj\u00e4nnen immunoglobuliinin kaltaisen domeenin l\u00e4heinen vuorovaikutus viidennen domeenin kanssa Syk:n sitoutumiseen viidenteen domeeniin. While thrombi are formed occasionally without the need to stop bleeding, a loose clot in the circulation may cause morbidity and mortality. Activation of platelets is a complicated process including an interplay of cell surface receptors and intracellular signalling networks. Spleen tyrosine kinase and the acting binding protein filamin are a part of this process.\nSpleen tyrosine kinase (Syk) is a cytosolic non-receptor tyrosine kinase, which is highly expressed in haematopoietic cells. Syk is known to have a crucial role in signalling. It plays some role in cellular functions like morphogenesis, cell growth, migration and cytoskeletal reorganization.\nFilamin is part of a family of three proteins that are products of distinct genes. They are large actin cross-linking proteins, but they are also known to interact with a wide variety of binding partners. Filamin serves as an integrator of cell mechanics and signalling.\nThe aim of the study was to confirm the binding between filamin and Syk and to determine the affinity of the binding. The special interest was to observe whether the filamin domain 4 has an effect on binding between filamin domain 5 and Syk, because a structure analysis has revealed that there is a novel type of domain-domain interaction in the structure of domain pair 4-5.\nInitial binding assay studies indicated a binding between filamin domain 5 and Syk. Filamin domain 4 had no apparent effect on the binding.
