Angiogeneesi tarkoittaa olemassa olevan kapillaarisuoniston kasvamista ja haaroittumista. Toimivien suonten kehittyminen on monimutkainen prosessi, joka vaatii tarkasti s\u00e4\u00e4delty\u00e4 yhteistoimintaa useiden stimuloivien ja inhiboivien signaalien kesken. S\u00e4\u00e4telymekanismi on niin monimutkainen, ettei sit\u00e4 ole onnistuttu j\u00e4ljittelem\u00e4\u00e4n keinotekoisesti. Liikunta on yksi harvoista mekanismeista, joka aktivoi angiogeneesin s\u00e4\u00e4telytekij\u00e4t toivotulla tavalla. My\u00f6s rasvaisen ruokavalion on havaittu lis\u00e4\u00e4v\u00e4n raajalihaksen kapillaaritiheytt\u00e4.\n\nT\u00e4m\u00e4n tutkimuksen tarkoituksena oli tutkia 20 viikon mittaisen rasvasy\u00f6t\u00f6n sek\u00e4 siihen yhdistetyn vapaaehtoisen juoksuharjoittelun vaikutusta angiogeneesin s\u00e4\u00e4telyproteiinien ilmenemiseen hiirten raajalihaksissa. Lis\u00e4ksi tutkittiin, miten rasvainen ruokavalio vaikuttaa maksimaalisen juoksutestin aiheuttamiin akuuttivasteisiin angiogeneesin s\u00e4\u00e4telyproteiinien osalta normaaliin ruokavalioon verrattuna. Pitk\u00e4aikaisvaikutuksia tutkittiin nelj\u00e4n ryhm\u00e4n avulla: kontrolliruoka (4% rasvaa) (n=10), kontrolliruoka+juoksu (n=10), rasvaruoka (60% rasvaa) (n=10) sek\u00e4 rasvaruo-ka+juoksu (n=10). Maksimaalisen juoksutestin akuuttivasteita tutkittiin nelj\u00e4n ryh-m\u00e4n (n=7/ryhm\u00e4) avulla: v\u00e4litt\u00f6m\u00e4sti testin j\u00e4lkeen lopetetut rasva- ja kontrolliruokaryhm\u00e4t sek\u00e4 kuusi (6) tuntia testin j\u00e4lkeen lopetetut rasva- ja kontrolliruokaryhm\u00e4t. Hiirten raajalihasn\u00e4ytteet homogenoitiin ja poolattiin. Angiogeneesin s\u00e4\u00e4tely-proteiinien m\u00e4\u00e4r\u00e4 analysoitiin Proteome prolifer array-menetelm\u00e4ll\u00e4.\n\nTutkimusjakson j\u00e4lkeen inaktiivisessa rasvaryhm\u00e4ss\u00e4 havaittiin angiogeneesi\u00e4 inhiboivien proteiinien laskua, mik\u00e4 on mahdollisesti rasvaruuan kapillaaritiheytt\u00e4 lis\u00e4\u00e4v\u00e4 mekanismi. Juoksuryhmiss\u00e4 havaittiin pitk\u00e4aikaisvaikutuksena ruokavaliosta riippumatta angiogeneesi\u00e4 stimuloivien proteiinien m\u00e4\u00e4r\u00e4n lis\u00e4\u00e4ntymist\u00e4, mik\u00e4 toiminee kest\u00e4vyystyyppisen kuormituksen kapillaaritiheytt\u00e4 lis\u00e4\u00e4v\u00e4n\u00e4 mekanismina. Akuuttivasteena maksimaaliseen kuormitukseen normaaliruokaisten ryhm\u00e4ss\u00e4 havaittiin sek\u00e4 stimuloivien ett\u00e4 inhiboivien proteiinien laskua, kun taas rasvaryhm\u00e4ss\u00e4 sek\u00e4 stimuloivien ett\u00e4 inhiboivien s\u00e4\u00e4telyproteiinien m\u00e4\u00e4r\u00e4 kasvoi l\u00e4ht\u00f6tilanteeseen verrattuna. Todenn\u00e4k\u00f6isesti molemmissa tilanteissa angiogeneesi tehostuu mekanismin ollessa erilainen. Tarkempien signalointireittien arvioiminen on t\u00e4ll\u00e4 menetelm\u00e4ll\u00e4 mahdotonta.
