Se koostuu toistuvista sokerimolekyyleist\u00e4 N- asetyyliglukosaminista ja glukuronihaposta. Hyaluronaani syntetisoidaan solukalvolla hyaluronaani- syntaasien (HAS) toimesta, joita nis\u00e4kk\u00e4ill\u00e4 on kolme isomuotoa (HAS1-3). Hyaluronaanin hajotus tapahtuu hyaluronidaasientsyymien (Hyal) toimesta. Hyaluronaanin p\u00e4\u00e4asiallinen solukalvoreseptori on CD44, jonka avulla hyaluronaaniketjut ovat sitoutuneet solukalvoon. Hyaluronaania esiintyy runsaasti etenkin sidekudoksessa ja kerrostuneessa epiteeliss\u00e4 kuten ihossa. Hyaluronaanin m\u00e4\u00e4r\u00e4n on havaittu lis\u00e4\u00e4ntyneen sy\u00f6viss\u00e4 ja t\u00e4m\u00e4 korreloi useasti potilaan huonon ennusteen kanssa. Hyaluronaanin lis\u00e4\u00e4ntyneen m\u00e4\u00e4r\u00e4n kasvainten stroomassa on havaittu tarjoavan hyv\u00e4n ymp\u00e4rist\u00f6n solujen jakaantumiselle ja migraatiolle. Usein t\u00e4m\u00e4 strooman hyaluronaani on kasvainta ymp\u00e4r\u00f6ivien solujen tuottamaa, jotka ovat aktivoituneen kasvainsolujen eritt\u00e4mien tekij\u00f6iden vaikutuksesta.\nMelanosyytit ovat ihon epidermaalisen kerroksen soluja, jotka tuottavat melaniini-pigmentti\u00e4. Melaniini suojaa ihon epidermaalisen kerroksen keratinosyyttej\u00e4 UV-valon haitallisilta vaikutuksilta. Melanooman kehityksess\u00e4 melanosyytit alkavat jakaantua yliaktiivisesti ja muodostavat hyv\u00e4nlaatuisen kasvaimen, joka my\u00f6hemmin muuntuu pahanlaatuiseksi melanoomaksi. Kasvainsolut toimivat vuorovaikutuksessa toistensa ja ymp\u00e4r\u00f6ivien solujen kanssa. Melanoomassa fibroblastit muokkaavat kasvaimen stroomaa ja s\u00e4\u00e4telev\u00e4t tulehdusvastetta. Melanooman invaasiolle paikallisen sidekudoksen hajotus on v\u00e4ltt\u00e4m\u00e4t\u00f6n prosessi. Fibroblastit tuottavat p\u00e4\u00e4asiassa n\u00e4it\u00e4 solunulkoisen v\u00e4litilan hajottavia proteiineja, matriksin metalloproteinaaseja, jotka ovat t\u00e4rke\u00e4ss\u00e4 roolissa kasvainsolujen invaasiossa ymp\u00e4r\u00f6iviin kudoksiin.\nMelanoomasolut eritt\u00e4v\u00e4t erilaisia kasvutekij\u00f6it\u00e4, jotka s\u00e4\u00e4telev\u00e4t fibroblastien aktivaatiota melanooman kehityksess\u00e4. Etenkin kasvainsolujen eritt\u00e4m\u00e4 TGB-! toimii kasvaimen kehittymisen alkuvaiheessa sit\u00e4 hillitsev\u00e4n\u00e4 tekij\u00e4n\u00e4 ja sen loppuvaiheessa aktivoivana tekij\u00e4n\u00e4. My\u00f6s PDGF ja VEGF ovat t\u00e4rkeit\u00e4 kasvutekij\u00f6it\u00e4 kasvaimen invasiivisen kasvun aikana, mik\u00e4 aktivoi angiogeneesin kehityst\u00e4. Tarkoituksena oli selvitt\u00e4\u00e4 hyaluronaanimetaboliaan liittyvien geenien ilmentymist\u00e4 qPCR-menetelm\u00e4ll\u00e4, proteiinitason muutoksia Western blot -menetelm\u00e4ll\u00e4 sek\u00e4 tutkia solujen morfologisia muutoksia immunohistokemiallisilla v\u00e4rj\u00e4yksill\u00e4. Tehdyt kokeet osoittavat C8161-melanoomasolujen eritt\u00e4v\u00e4n kasvuliuokseensa tekij\u00f6it\u00e4, jotka aktivoivat fibroblasteissa hyaluronaanisynteesi\u00e4. Etenkin seerumin sis\u00e4lt\u00e4v\u00e4 kasvatusliuosk\u00e4sittely lis\u00e4si huomattavasti fibroblastien Has2:n mRNA:n ilmentymist\u00e4, mik\u00e4 vahvistettiin my\u00f6s proteiinitasolla. K\u00e4sittelyll\u00e4 ei havaittu olevan vaikutusta CD44:n ja Hyal2:n mRNA tasoihin.
