Vaikka faagien genomeista suurin osa on alle 200\nkiloem\u00e4sparia, my\u00f6s suurempia jumbofaageja sek\u00e4 megafaageja on eristetty tai\nkoottu metagenomiaineistosta. Megafaageja ei ole kuitenkaan saatu eristetty\u00e4,\nmink\u00e4 vuoksi niiden biologia tunnetaan huonosti. T\u00e4ss\u00e4 ty\u00f6ss\u00e4 tutkittiin\ntiett\u00e4v\u00e4sti ensimm\u00e4isen megafaagi-isolaatin, FKy-1:n vuorovaikutusta\nFlavobacterium sp. B222 -is\u00e4nt\u00e4bakteerin kanssa. Tutkimuksessa tarkasteltiin\nfaagin infektiivisyyden pysyvyytt\u00e4 eri olosuhteissa. Huomattiin, ett\u00e4 faagin\ninfektiivisyys laskee kloroformin vaikutuksesta, mutta se s\u00e4ilyy\nhuoneenl\u00e4mm\u00f6ss\u00e4 j\u00e4rvivedess\u00e4 yht\u00e4 hyvin kuin +4 \u00b0C:ssa kasvatusalustassa.\nT\u00e4m\u00e4n lis\u00e4ksi m\u00e4\u00e4ritettiin faagin elinkierto sek\u00e4 tutkittiin faagin\nvuorovaikutusta is\u00e4nt\u00e4bakteerin kanssa j\u00e4rvivedess\u00e4 ja mediassa. They are found in a wide range of\necosystems. Most of the phages are less than 200 kilobase pairs, but larger\njumbophages and megaphages have been found. While megaphages have been\ndetected in metagenomic data, no isolates have been obtained. Because of that,\nbiology of megaphages remains poorly understood. In this study, we analysed\nthe interaction between the first known megaphage isolate, FKy-1, and its host,\nFlavobacterium sp. B222. We followed the stability of the infectivity of the\nmegaphage in different conditions. It was found that chloroform causes\ninfectivity loss on the phage. Also, we noticed that phage infectivity remains as\nstable in lake water at room temperature as in medium at +4 \u00b0C. We determined\nthe life cycle of the FKy-1 phage and observed differences in the phage-bacteria\ninteractions in lake water and in media. Stages of the phage life cycle was\ndetermined by electron microscopy from thin-slice-samples. Significant changes\nin the bacterial cell counts were observed approximately 3 hours after infection.\nAlso, lake water can be used as a growth medium, but it results in lower viral\nparticle and cell counts. In addition, it was found that lake water induces\nmorphological changes in the bacterial cells. This finding indicates that phagebacteria\ninteractions can be studied in lake water, but these limitations must be\nconsidered.
